In this paper, a method-immobilized antibody modified with self-assembled monolayer (SAM) has been described by means of covalent binding with 1-ethyl-3-[3-(dimethylamino)propyl] carbodiimide hydrochloride (EDC) or glutaraldehyde. The preparation of the immunosensor is described as followed: 1.The electrochemical behavior of gold planar electrode ,silver disk electrode, gold disk electrode has been investigated. They were used to fabricate the immunosensor, respectively. It shows that the immunosensor using gold planar is more sensitive and more stable. 2. The electrochemical behavior of three different SAM [SH(CH2)10COOH ,EG6OCH2COOH and cysteine ]electrodes has been investigated.It shows these three thiol were successfully immobilized on the gold planar because the potential change great.Different thiol were used to fabricate the immunosensor, It shows that the immunosensor using SH(CH2)10COOH has a larger detection range and a better linear relationship.  3. The suitable temperature and pH to prepare of the immunosensor recorded in literature is 30℃ and pH=5.5.  4.The response of the immunosensor to mouse IgG at different temperature and pH has been determined. It shows the suitable temperature and pH for immunoreactions is 30℃and pH=5.5. 5.The characteristics of the immunosensor using SH(CH2)10COOH have been investigated in detail. It can detect the target molecule in the range of 0.4-12ng/mL. The detection limit to mIgG is 0.4ng/mL, the correlation coefficient is 0.9912,the response time is 12 min, the immunosensor can be regenerated using 0.04mol.L-1HCl and its lifetime is 16 days. 6.The characteristics of the immunosensor using cysteine have been investigated in detail. It can detect the target molecule in the range of 0.05-5ng/mL, The detection limit to mIgG is 0.05ng/mL, the correlation coefficient is 0.9931,the response time is 14 min, the immunosensor can be regenerated using glycin-HCl buffer (pH2.3).
