Lotus leaves , dry leaves of Nelumbo nucifera Gaertn. , had the efficacies of losing weight, reducing fat and decreasing hypertension ,etc. The components in Lotus leaves include alkaloids , flavonoids and organic acids ,etc. In modern research, scientists had found that lotus leaves had the activities of stop bleeding, decreasing blood pressure, reducing weight and resisting arrhythmia and so on. Alkaloids were known of the main bioactive ingredients in it. Flos Daturaes, dry flowers of Datura metel L., had the efficacies of relieving cough, easing pain and releasing from hyperkinesias etc. Scopolamine and anisodamine are the main bioactive ingredients in Flos Daturaes.Part one: The research of alkaloids in lotus leavesFirstly, the studies of lotus leaves and the applications of macroporous resin in the separation of effective components in alkaloids were summarized. Secondly, We established the method of determining nuciferine in lotus leaves by HPLC in order to ensure the well quality of technics, investigated the method of extracting alkaloids from lotus leaves with ethanol, and enriched alkloids of lotus leaves with macroporous resin. One macroporous resin was selected from nine macroporous resins. With the content of nuciferine as the marker, the optimized adsorpting conditions including the ratio of dia-height, the dynamic adsorpting capacity, the concentration of sample, the velocity of adsorption and the leak curve were studied. The research also optimized the conditions of elution such as the kind of eluting solvents, the velocity of elution and the eluting curve. On the base of above-mentioned optimized conditions of adsorption and elution, the repetition of the technics was studied with the fineness of nuciferine as the marker. With the optimum technological parameters of the enrichment process, the study provided a new technology which could enrich alkaloids in lotus leaves.Part two: The study of the determining method of Scopolamine in Flos DaturaeThis paper summarized the studies of Flos Daturae and the determinating methods of Flos Daturae. The method of determining Scopolamine in Flos Daturae by HPLC was established, and the choices of distilling solvents and the optimum hromatographic condition were investigated. This method was simple, accurate, higher sensitivity and the reproducibility was good, so the method could be used for the qualitative study of Flos Daturae.
