| Supercritical carbon dioxide fluid extraction of colchicine from thebulbs of lily was studied and the extraction technological conditions wereoptimized by vertical-method. The contents of colchicine in extracts weredetermined directly by high performance liquid chromatography (HPLC).The result showed that at 40, ISMPa and with ethanol as pre-soaker,the extraction efficiency is the highest after lily powder was alkalified byammonia liquor. Under the given conditions, colchicine concentrationincreased greatly from 0.049% in natural sample to 6.4% in extract with- supercritical fluid extraction(SFE). The comparison of SFE and organicsolvent extraction suggested that the former is a quick, safe, non-toxicand environmental friendly method with higher yield and selectivity. Sosupercritical fluid extraction of colchicine from the corms of lily ispracticable.Colchicine was also successfully extracted from the waste water which was produced in the course of lily food production.Pure colchicine was prepared from the extracts through thin-layer chromatography and its proper procedure was found. |
