A fine procedure for the isolation and purification of insulin from small quantities of porcine pancreas is established. Radioimmunoassay is the method of detection for insulin. The steps involve extraction with acidified ethanol, removal of alkaline and acidic protein, isoelectric precipitation of the insulin with ZnCl2, 23%(w/v)sodium chloride is added to get the crude insulin. Further more, on QAE-Sephadex A-25 with a Tris-HCl buffer and 0.05mol/L HCl, insulin was eluted as a single peak. |
