SUMMMYChinese herbal drugs are the gems of the five thousands years old civilization of our country. According to statistic, the export of the Chinese herbs produced in China is only 3% of the trade of herbal drug of the world. The reason for which is lack of modern analytical methods for the effective components extraction, the quality control, and the pathology investigation. Therefore, how to develop the advanced analytical methods for the research of research Chinese traditional medicine is the important task of the analysis of medicine in the future.Salivia Miltorrhrza, a Chinese traditional herbal drug, is used for treatment of many diseases, such as cardiodynia, myocardial infarction, pyrogenic infection, hypochondriac pain, stomach ache dysmenorrhea, trauma with blood stasis. It contains waters soluble phenols and fats soluble tanshinones. According to pharmacological tests, in the waters soluble phenols, protocatechuic aldehyde and protocatechuic acid are the effective components of healing of articardiac angina, which were found to possess the great effects on the improvement of coronary arteries flow and the inhibition of the aggregation of platelets caused by ADP (adenosine diphosphate). And in the fats soluble tanshinones, Tanshinone IIA not only is applied to resist oxidation, counterplea and diminish inflammation, but also has obvious effect on cure of cancer. Some analytical methods for determination of protocatechuic aldehyde, protocatechuic acid and Tanshinone IIA are developed in this thesis.Section 1 Separation and determination of Protocatechuic Aldehyde and Protocatechuic Acid in Salivia Miltorrhrza by Capillary Electrophoresis with Amperometric DetectionCapillary electrophoresis with amperometric detection was applied to separate and determine protocatechuic aldehyde and protocatechuic acid in Salivia Miltorrhrza preparations in this paper. The electrode used was 0.3mm diameter carbon disk electrode fixed in a wall-jet amperometic detection. Under the optimum condition, the two analytes were separated completely within 8 min. This method has been successfully applied to-3-monitor these two components in real samples such as Salivia Miltorrhrza and its two traditional Chinese medicinal preparations.Section! Study on the electrochemical behavior of protocatechuic aldehyde at the glassy carbon electrode and its applicationThe electrochemical behavior of protocatechuic aldehyde (3, 4-dihydrobenzoic aldehyde ) was investigated by linear-sweep voltammetry , cyclic voltammetry differential-pulse voltammetry at the glassy carbon electrode. The peak current showed a linear relationship with concentration in the range of 0.25ug/mL-2.5 ug/mL and 5 ug/mL-25 ug/mL by differential-pulse voltammetry. The detection limit is 0.10 ug/mL. The mechanism of the electrode reaction has been discussed. This method has been employed satisfactorily for the determination of protocatechuic aldehyde in serum by standard addition.Section2 Separation and determination of waters soluble and fats soluble components in Salivia Miltorrhrza by Micellar Electrokinetic Capillary ChromatographyA high performance method has been developed for separation and determination of waters soluble (protocatechuic aldehyde and protocatechuic acid )and fats soluble components (Tanshinone II A) in Salivia Miltorrhrza by Micellar Electrokinetic Capillary Chromatography(MECC). At 25癈 and 15kV applied voltage, the three compounds were baseline separated within 14min in 20mmoL/L NaH2PO4-Na2B4O7 buffer (pH8.63)including 60mmoL/L SDS-15 mmol/L SC at the detection wavelength of 214nm. This method has been successfully applied to analyze these components in real samples, such as Salivia Miltorrhrza and its three traditional Chinese medicinal preparations and simulated serum sample.
