This thesis studies the analytical methods of determination of Leucogen and Hydroxymethylnicotinamide by Ultraviolet and Atomic Absorption Spectrscopy. Three determinate methods of Leucogen and a new method of Hydroxymethylnicotinamide by first-order derivative Ultraviolet spectrophotometry are reported.The methods have been applied to determine the samples and satisfactory results were obtainded.It includes five parts as follows:Part Ⅰ: Review. A review of the application of Ultraviolet spectrophotometry and atomic absorption spectroscopy in pharmaceutical analysis is given.Part Ⅱ: Determination of Leucogen by indirect Ultraviolet Spectrophotometry. The method of determining Leucogen indirectly by Ultraviolet Spectrophotometry first is reported.It is based on the strong absorption in 220nm when Leucogen is resolved in alkaline solution,determine the amount of Leucogen according to it's absorptivity in 220nm.The relative deviation between this method and titrimetry is less than 1.0%.The linear range is 10.0~100.0μg·mL-1, ε is 1.13×104 L?mol-1?cm-1 and the recovery is 103~105%.This method is simplier and more rapidly than titrimetry used in pharmaceutical factories at present.Part Ⅲ: Determination of leucogen by indirect atomic absorption spectroscopy. The method of Leucogen indirectly by atomic absorption spectorscopy first is reported.It is based on the reaction of cysteine that is produced by Leucogen'hydrolytic cleavage with Cu2+ in alkaline medium,determine the amount of Cu2+ in the supernatant solution by AAS,and then determine the amount of leucogen.The linear range is 10.0~100.0μg·mL-1,the recovery is 98.5~102%.When the method has been applied to determine Leucogen with good result.Part Ⅳ:Determination of Leucogen of Leucogen tablets indirectly by Flow Injection-Spectrophotomertry.The method of Leucogen indirectly by Flow Injection-Spectrophotomertry first is reported. The compound formed by the Leucogen'hydrolytic cleavage in strongly alkaline medium is spectrophotometrically determined at λ300nm to obtain the amount of leucogen. The determination frequency is 60 times per hour.The linear range is 10.0~200.0μg·mL-1,and the recovery is 101~102%. The method has been applied to determine Leucogen tablets with satisfactory results and is simplier and more rapidly than titrimetry used at present.Part Ⅴ:Study on the mothod of determination of Hydroxymethylnicotinamide by first-order derivative Ultraviolet spectrophotometry. The UV spectrophotometry of Hydroxymethylnicotinamide solutions in water, 0.01mol·L-1 NaOH and 0.01mol·L-1 HCl is studied and a new determinating methid of Hydroxymethylnicotinamide named first-order derivative UV spectrophotometry (top~zero) first is reported. When this method is applied to determinate Hydroxymethylnicotinamide,it can obtain good result with a wide range and a little relative error.
