This paper presented double wavelength thin layer chromatography scanning,a method for the quantitative determination of chemical composition in Chineseherbal medicine. The analytical techniques to determine the content of Baicalin inRadix Scutellariae and some Chinese medicines containing Radix Scutellariae weresummarized.A new version of the internal standard method in quantitative analysis wasfound. One of the advantages of the internal standard method is that the result of theanalysis is not affected by variations in the volume of sample,so that the method iswidely applied in gas and high performance liquid chromatography. However, themethod is not often used in thin layer chromatography for its unproportionalrelationship between the amount of the determined substance and the signal from anoptional detector. A new version of the internal standard method in thin layerchromatography was found. The new method overcame the disadvantages of theconventional internal standard method and enabled to enhance the accuracy andprecision of quantitative analysis. The content of Baicalin in Radix Scutellariae,Tongxuan Lifei pill, Huaijiao pill, Mengshi Guntan pill and Gegen Qinlian capsulewere determined by this new approach. The eluent was C6H6:CH3COCH2CH3:CH3OH:HCOOH(9:2:1.5:0.5).The fixed phase was polyamidation film.Doublewavelength reflection zigzag scanning was performed. The results were reliable andthe average recovery was satisfying. In addition, the quantitative determination of Hesperidin in Baohe pill,Marenpill etc.were described in this paper. The eluent was EtOAc: CH3OH:H2O(10:2:13);The fixed phase was Silica G. Double wavelength reflection zigzag.scanning was performed. The regression equation is Y=11822x-1194.36,r=0.9992.The results were reliable.
