Objectives: The research is according to the clinical prescription of the powder Xanthium sibiricum,to prepare compound Flos Magnoliae naristillae. The preparation has the function of Dispelling wind heat and crelieving stuffy nose. It was mainly treats the allergic rhinitis. In this experiment, the extracting method of the volatile oil and extract of Flos Magnolia flower were prioritized. The method of thin layer chromatography(TLC) and high performance liquid chromatography(HPLC) were used to qualitative and quantitative analysis the main components in order to control the preparation quality. Meanwhile, to the model of allergic rhinitis in rats were established to observe the pharmacodynamic action of anti allergic rhinitis of compound Flos Magnoliae naristillae.Methods:By steam distillation extraction method, this paper mainly studies extraction time,solid-liquid ratio and extraction times of three factors, each factor selected three levels and through the orthogonal design experiment to determine the optimal process parameters of yield of Flos Magnoliae volatile oil. By reflux extraction method,ethanol concentration,solid-liquid ratio and extraction time as factors, each factor selected three levels and through the orthogonal design experiment to determine the optimal process parameters of the yield of dry extract and the content of Mulan ester element. Qualitative identification of preparation was by TLC method, HPLC method was used to determine the content of Mulan ester element of Flos Magnoliae. The rat model of allergic rhinitis was established by egg albumin in order to observe the inhibitory effect of compound Flos Magnoliae naristillae on rats with allergic rhinitis.Results : The optimization extraction method of volatile oil of Flos Magnoliae is the solid-liquid ratio 1:6, soaking time12 h, heating extraction 6h, extraction times 4. The optimization extraction method of extract of Magnolia is ethanol concentration85%,solid-liquid ratio 1:10, extraction time 1.5h, extraction times 3 times. The established TLC method and HPLC method are rapid, simple and reproducible, and can be used for quality control of Compound Flos Magnoliae naristillae. Pharmacodynamic test showed that compound Flos Magnoliae naristillae is without obvious mucosa stimulation, cause allergic reactions and obvious toxic reaction, which can effective against allergic rhinitis rats induced itching,sneezing, runny nose symptoms by egg albumin generated by nasal.Conclusions : The extraction, preparation technology of compound Flos Magnoliae naristillae are simple, stable. The quality standard of the preparation are accurate, sensitive,reproducible, and can be used for quality control of the preparation. Compound Flos Magnoliae naristillae can effectively inhibit the nasal inflammation and has the efficacy of anti allergic rhinitis.
