With the development of food industry and the change of people consumption idea, the health products of natural activity have become the research hot spot,and the flavonoids with its natural, quick effect, highly active, have been pay more and more attention by people. Based on this, this article on the one hand,has explored the extraction methods of flavonoids in sensitive plant seeds, on the other hand has established the separation collection methods of flavonoids, as well as has separated four kinds of flavonoids of common by HPLC.This paper will be divided into four chapters, the main content is as follows:In chapter 1: Summarize the method of extraction and separation of flavonoids,and describes the background, research contents, and innovation.In chapter 2: Establish the ultrasonic extraction method of total flavonoids in sensitive plant seeds.Investigate the effects of ultrasonic extraction rate in solvent composition, ingredients ratio, ultrasonic time, ultrasonic temperature and soaking time,at the same time,determine the content of total flavonoids while using Na N02-AI(N03) 3 colorimetry with rutin as the reference.Carry on the orthogonal experiment on the basis of single factor,obtained the optimum extraction: ethanol concentration is 60%, the ratio of ingredients is 1: 8, ultrasonic temperature is 40 ℃- 44 ℃, ultrasonic time is 50 min.This method is simple, short extraction time, and high extraction efficiency.In chapter 3: Establish a purifying method of flavonoids in sensitive plant seeds with macroporous adsorbent resin Diaion HP-20. Under the existing conditions of the purification process,discuss the effect of eluent volume fraction and amount of eluent for separation,and monitor by HPLC.Obtaine that :when eluent concentration is 100% ethanol, eluent amount is 3BV, the eluent volume fraction is the highest, and flavonoid content is the highest;Collect and detect the samples, respectively, by means of a preparative high performance liquid chromatography, under optimal separation conditions.In chapter 4: Establish a separation method by HPLC for the four common flavonoids at the same time. Investigate the effects of wavelength, sample concentration,flow rate, column temperature and the mobile phase composition, for the separation. Obtaine the optimal separation conditions: Column is C18 column(4.6 × 250 mm, 5μm); mobile phase is v( methanol):v( water)= 60: 40; detection wavelength is 260 nm, flow rate is 1.5 m L / min, the column temperature is room temperature. And the lowest detection limit of four substances are detected, namely: 240μg / L, 30.0μg / L, 17.5μg / L, 9.12μg / L, and the recoveries are between 86.4% ~ 110.6%. The method has high sensitivity, short cycle analysis and has great reference value for the qualitative detection of flavonoids.
