| Pomegranate has high value of comprehensive utilization of nutrition, health, beauty, in recent years, pomegranate cultivate and product industry have increased greatly, but a lot of pomegranate seeds( about20% ~ 30% for pomegranate) were abandoned, resulting in a waste of resources, pomegranate seed oil is about 20% in pomegranate seed, the protein of the residue(14.74%)can also be utilized as a kind of protein resource. Taking pomegranate seed of Xinjiang Kashgar Oasis fruit limited liability company after juicings as raw material in this study, the ultrasonic assisted extraction of pomegranate seed oil, then get the optimum of extracting pomegranate seed protein preparation, and researched the functional properties of pomegranate seed protein, the results are as follows:(1) With the orthogonal experiments. the optimal extracting conditions are liquid-solid ratio1: 12 g /mL, ultrasonic power 200 W, ultrasonic treatment time 50 min and ultrasonic treatment temperature 40℃. the best yield is 15.16%.(2) Xinjiang pomegranate seeds oil have excellent physicochemical properties; The GC-MS result shows that, the main fatty acids components were: palmitic acid 6.24%,linoleic acid 14.64%, oleic acid 11.11%, punicic acid 43.46%, stearic acid 3.97%, 9,12,15-octadecatrienoic acid 11.31%, 6,9,12-octadecatrienoic acid 9.28%. degree of unsaturation is high, The varieties and source, extraction methods of pomegranate have a certain effect to fatty acid content;(3) Pomegranate seed protein was comoposed of 78.13%Glutelin,5.92%Albumin,5.92%Gobulin,2.91%Prolamin,9.55%Compound protein.Glutelin is the main.(4) The optimal extraction using the response surface methodology of pomegranate seed protein were as follows:p H 10.3,material- to-liquid ratio1:33(g /mL),temperature 46.8 ℃, time 40 min,resulting in an high extraction yield of 80.14%,the yield of pomegranate seed protein was 12.77%, the purity was 81.43%the pomegranate seed protein pI=4.20;(5)The optimal ultrasonic extraction using the response surface methodology were as follows :pH 10.2,material- to-liquid ratio 1:32(g /mL),ultrasonic temperature 39℃,ultrasonic time 31 min and ultrasonic power200 W,resulting in an high extraction yield of 85.94%, the yield of pomegranate seed protein was 14.20%, the purity was 79.86%(6) Pomegranate seed protein is composed of 17 kinds of Amino acid by ultrasonic extracting,eight of which are essential for the body accounted for 34.83% of total amino acids. The first limiting amino acid is lysine,and the profile of Amino acid is 38, pomegranate seed protein is not quality protein;(7) The solubility, foaming and foam stability, emulsifying capability and emulsion stability of Pomegranate seed protein are affected greatly with pH increasing, as "V" trend, the functional properties are poor at p I of protein.Functional properties of pomegranate seed protein with the increase of the temperature have increased, but when the temperature is higher than 60℃, the functional properties were decreased;the solubilityand emulsifying properties,foaming properties water holding,oil holding of pomegranate seed protein are affected by the increase of NaCLconcentration showed "∧", at the 0.6mol/L NaCL concentration,the functional properties often are good.The addition of sucrose is in favor of pomegranate seed protein,when sucrose concentrations is 4%, functional properties are the best, protein foaming properties, emulsifying properties of pomegranate seed increased with the increase of protein concentration, protein concentration(CMC) is about 0.6%. |
