Vitamin A palmitate is one of commonly used vitamin A products. Vitamin A palmitate can help maintain normal visual ability and participate in various metabolic activities to maintain the body health, which can be adopted in food, cosmetics, and medicine industries as additives. Currently, Vitamin A palmitate is produced by chemical method in commercial process. However, compared with chemical synthesis, enzymatic synthesis of Vitamin A palmitate has the advantages of mild condition, simple process, less energy consumption and being environmental friendly, which has application prospect. But the synthetic process of laboratory at present has high cost and security problems caused by the presence of organic solvents. These problems restrict the process of further enlarge.This paper aims at reduce production cost and discover suitable forms of scale up. This paper focus on the control of solvent synthesis process to reduce the cost of the lipase and solvent.at the same time, develop a solvent free synthesis process. The two process were compared.In the solvent system, the substrate concentration is increased by the optimization of extraction conditions. The production costs of solvent and enzyme are further reduced by biological basis dehydrant and diatomite immobilized enzyme. The results shows that the biological basis dehydrant has a better effect of water removal than traditional dehydrant, with a water absorbent rate of 10g water/g agaric powder. Enzyme amount can be reduced by 70% using diatomite immobilized enzyme, and the catalysis effect can be raised by 1.6times. The enzyme can be reused for 12 batches in 3L stirring reaction. The enzyme cost of producing per kg products can be reduced from 6.7 yuan to 2.4 yuan, and the solvent cost of producing per kg can be reduced from 25 yuan to 12 yuan.An environmental friendly solvent free process is developed. Vitamin A acetate and ethyl palmitate are utilized as substrates. Viscosity of the substrates and water control are studied. The conversion can be increased to 80% by using diatomite immobilized enzyme, optimization of reaction conditions, and section reaction. Scale up is achieved to verify the possibility. Enzyme can be reused for 3 batches. Methanol is utilized as an extraction agent. The purity of Vitamin A palmitate can reach 94%.
