1,3- propanediol (1,3-PDO) is a new kind of chemical raw materials,which has very good prospects in the synthesis of PTT,In today’s mainly by chemical synthesis, biological methods because of its various advantages and has received the widespread attention.This article mainly discusses three main items:First,Through the 5L fermentation experiment screening good strains which show better in the shake flask,we obtain two strains named kp-fug and kp-13-fug; For the two strains by using DPS design to optimize the fermentation medium and the loading of the nitrogen source and glucose matter,the 5L fermentation level were 81.7g/L(1,3-PDO), and 22g/ L(2,3-BDO), and diol conversion rate above 0.63mol/mol; Using calcium hydroxide partly replace potassium hydroxide in alkali method, the final 5L fermentation system product level was 87g/L, the main by-product of 2,3-butanediol concentration was 29.9g/L, diol conversion rate above 0.74mol/mol and reduces the cost of fermentation.The project cooperation with Ruixing chemical plant in shandong, completed the annual output of 2000 tons of production plant construction, and entered into trial operation stage. Preliminary experiments in a factory, fermentation strains, concentration of antibiotics and optimum control condition of factory equipment has been determined.we designed a fine experiment scheme for the problems arising from the factory fermentation,and using laboratory conditions to solve one by one,finally found that the factory of glycerol and tap-water excess sodium and chloride ions which in klebsiella peneumoniae inhibit metabolic pathways.After sure the reason we put forward the corresponding solution,for the smooth production of the late we accumulated a large number of experimental data.Finally, we examine the change of metabolic pathways When bacteria lives in different dilution rates and different feeding sugar concentration using continuous fermentation means. In the base of continuous fermentation,Regulating different dilution rates and different feeding sugar concentration, we simulate the various stages of fermentation.In order to adjust the fermentation process under the effect of glucose and glycerol,We explorer the optimal glucose concentration of cell growth.
