The selective isolation and detection of protein targets from complex samples is of great significance in key life science disciplines. The magnetic materials allow the rapid development of the protein separation technology. Its fast and efficient separation attract so many researchers that more and more studies tend to choose magnetic materials as a carrier for the separation of proteins. Numerous methods have been developed to tackle the separation issues through specific interaction between protein tag and surface ligands. The polymerization is applied for modifying the magnetic carrier in this paper and we focus on its ability in protein separation.In this study, we integrated the advantages of magnetic materials and polymerization methods to build a powerful and convenient technique for heme proteins separation. With free radical polymerization, the polymer coating possessing the imidazole ring was grafted onto the surface of magnetic particles.The morphology, structure and composition of the materials were characterized by transmission electron microscopy, Fourier transform infrared spectroscopy and thermogravimetric analysis. The resulting polyvinyl imidazole modified magnetic micrometer-sized particles (PVIM-MMPs) showed a strong magnetic response to an externally applied magnetic field, a remarkable adsorption capacity (6.96 g/g) and excellent specificity (compared with non heme protein-Lysozyme) and good recyclability toward model heme protein-hemoglobin. Also a serious of separating behaviors of PVIM modified magnetic nanoparticles (PVIM-MNPs) were investigated, and through a series of characterization tests, the results show that the polymer-modified particles exhibit better adsorption capacity. This study provide a choice for the carrier of future separation of biological macromolecules.Above all, we established a method for preparation of a polymer modified material, and it can separate heme protein selectively with simple pre-treatment, high separation efficiency and easy operation. This technology will help to provide more ideas for remove technology of high-abundance proteins, and enrichment of low-abundance proteins.Especially it is of great importance for the separation and purification of heme proteins.
