Inonotus obliquus (Ft.) Pilat has been known for hundreds of years as a medicinal fungus in Russian Federation and Siberia.I. obliquus has the activities of the antioxidant, anti-tumor, anti-AIDS virus, anti-sudden change, stimulating immunity, reducing the blood sugar and so on. On the purpose of high yield of fermented mycelia, and intracellular and extracellular polysaccharide, the Ⅰ. obliquus from the East China Pharmaceutical Group Limited Company was as the original strain. Optimization of agar slant, degeneration and rejuvenation of Ⅰ. obliquus was studied. According to the index of mycelium dry weight of Ⅰ. obliquus, the fermentation technology of shake flask and 700 L-fermentation tank were also preliminary researched. Moreover, the effect of flour of Ⅰ. obliquus on streptozocin (STZ) induced diabetic mouse was observed. The results were as follows.1. The best suitable for the cultivating the Ⅰ. obliquus was modified PDA Ⅱ (potato extract 20 g/L, Glucose 20 g/L,. MgSO4 1.5 g/L, KH2PO4 3 g/L, thiamine VB1 0.1 g/L, agar 15g/L) agar slant, on which the mycelium of the strain was to be white, fuzzy and full.2. The optimum fermentation conditions of shake flask were as follows: temperature 28℃, rotary speed of shaking flask is 150 r/min, inoculation amount 3%, the time of transference from the level 2 seed broth to the shaking flask fermentation is the 7th day, and the fermentation period of shake flask is 8 days. The optimum fermentation medium was as follows:glucose 3%, corn meal 1.5%, soybean flour 2%, arginine 0.1%, MgSO4 0.15%, KH2PO4 0.3%, pH 6.0. Under the optimum fermentation medium, the yield of mycelium dry weight of I. obliquus was 23.3 mg/L. In addition to this, the fermentation technologies of pilot scale test were inoculation amount 10% and 8 days’ fermentation periods of both level 1 and 2. Under the fermentation conditions above, the yield of extracellular and intracellular polysaccharide of ′. obliquus were 17.93 mg/L and 56.19%, respectively.3. The activity of flour of Ⅰ, obliquus on STZ induced diabetic mouse was tested. In the meantime, the experimental evidence for Ⅰ. obliquus as a new kind of fungus drugs which could prevent and cure diabetes. The results were as follows:the fasting blood glucose (FBG) of drug group mouse were degraded significantly (P<0.01); the low dose (200 mg/kg) group of Ⅰ. obliquus group don’t degrade the the FBG, but the high dose (400 mg/kg) group could reduce the blood sugar of the tested mouse (P<0.05). In other words,400 mg/kg flour of Ⅰ, obliquus was able to degrade the level of blood FBG of STZ induced mouse.
