Chinese have successfully resolved the problem of feeding. Thus in our daily life, wecare more about food safety and hygiene than how to eat our fill. But we are constantlytensed due to food hygiene events. The number of bacteria in food is an importantindicator to judge food hygiene and freshness. The number of bacteria in food productionsites can also reflect whether the food production environment met the required standards.The bacterial number detection is becoming more and more important for food safety.The traditional bacteria counting method have optical microscopy directly countingmethod, fluorescent microscope counting method, and plate counting method method.The traditional bacteria counting methods are easily affected by the condition of testingequipment, the complex degree of detection method and the quality of testing personnel.These counting method can not detect the sample in real-time.For example, the optical microscopy directly counting method need testing personnelto extract samples at the scene and take sample to the lab, then observe the sample underthe microscope and count the number of bacteria. The optical microscopy directlycounting method require testing personnel with high quality and also should haverelevant biological technology background to complete the detection. The detectionresult is easily effected by human factors and the sample can be detected only when itwas brought back to the laboratory. It takes a long time to get the number of bacteria,which apparently restricts the applicable occasions and scope. The traditional detectionmethods do not meet the requirements of on-site rapid detection. Fluorescencemicroscopy counting method and other laboratory counting method also have all kinds ofproblems. So these methods do not meet the requirements of rapid bacterial detection.In order to rapidly bacteria detection, we propose colony rapidly detection system. Itwas based on ATP bioluminescence, the latest photoelectric conversion and electronic technology. ATP is short for Adenosine Triphosphate which is the energy supply material forbacterial cell and microbial cells. Every bacterial cell contents the roughly same amountof ATP. The amount of ATP is linearly proportional to the number of bacteria. Theamount of ATP can indirectly reflect the number bacteria. However, ATP is inside thecells thus it can not be detected directly only when it’s dissociated from the bacterial cells.When catalysed by oxygen ion, magnesium ion and luciferase, ATP reacts withfluorescein and can emit fluorescence, which is linearly proportional with the amount ofATP, i.e., the fluorescence intensity can reflect the amount of ATP. A/D conversion willconvert analog voltage signal into digital voltage signal which will be sent to (MCU) orcomputer. Then signal can be processed through computer.The bacteria detection system based on ATP bioluminescent has merits whichtraditional detection method can not compare with. Firstly the test time is shortter, andthe number detection of bacteria can be finished in minutes. The system is portable andadapt to the demand of field testing. Thirdly the system is easy to operate and the testerscan operate the detection system without professional training. On the accuracy andrelevance, this detection system can reach more than90%of the traditional method. Ifthe detection system combined with traditional methods, the detection accuracy can befurther improved.ATP bioluminescent bacteria detection system has widely range of applications. It notonly can be applied to food hygiene but also can be applied to healthcare. If the detectionsystem combined with other medications, it can be used to monitor the sensitivity ofcancer cell drug sensitivity etc. Also it can be applied to life sciences, ecological sciences,natural environmental monitoring, water quality monitoring,ect.The detection system combined with other disciplines can greatly broaden itsapplication scope. It has important reference and guiding significance for scientificresearch. Its further application and scientific value need to explore.
