| Probiotics play an important role in maintaining the gastrointestinal health ofhost, balancing and improving the immunity and modulating the ways of bodyglucolipid metabolic. To some extent, modern life pressure (eating disorders, high-fatfood intake, antibiotic abuse, excessive alcohol consumption and the environmentalpollution) causes the number of probiotics significantly decreased, intestinal microbialimbalance and induces a variety of chronic disease and makes human body in a stateof sub-health. Appropriate supplementary probiotic preparations could improve thebody’s immune system, which prevents and treats of some chronic diseases. The highquality probiotics preparations containing high number of living bacterium, andpossessing additional functional health can better meet the needs of people for greenhealth food. This paper studied the key technologies of the proliferation of probioticsand the development of probiotics preparations. The research and main results are asfollows:Probiotics separation and molecular identification based on16s rDNA. Somebacteria strains were isolated and obtained from12kinds of probiotics medicinescontaining different bacteria strains. The11strains of probiotics were identificationby the molecular identification based on16S rDNA. The11strains of probiotics werelactobacillus rhamnose, lactobacillus rhamnose SD-5217, lactobacillus plantarum,lactobacillus salivarius, lactobacillus casei, Lactobacillus fermentum, Lactobacillusdelbrueckii subsp. bulgaricus, Streptococcus thermophilus, Enterococcus faecium,Bacillus coagulans and Bacillus subtilis, respectively.The influence of plant source additives on lactobacillus growth in vitro wasconducted. This experimental study of7kinds of plant source additives are: threekinds of prebiotics (fructo-oligosaccharide, Isomalto-oligosaccharide andGalacto-Oligosaccharides) and three kinds of plant extracts (Mulberry leaf extract,Cordyceps Sinensis extract, Ganoderma lucidum extract and rutin), respectively. Wefound that six kinds of plant source additives could promote the proliferation oflactobacillus rhamnose in vitro. When added individually5%(m/v) of prebiotics or5 mg/mL of plant extract into the cultivation system of lactobacillus rhamnose in vitro,to some extent, the plant source additives promote the proliferation of lactobacillusrhamnose and raise the total bacteria. But the influence of the lower concentration ofbotanical extracts on lactobacillus rhamnose is not obvious. The7kinds of plantsource additives had no inhibitory on the growth of probiotics. Therefore, theseadditives could add into the probiotics and may, to some extent, promote the numberof intestinal probiotics.To optimize the fermentation medium of probiotics, we use the response surfaceanalysis and optimize nine experimental factors affecting the growth ofbifidobacterium breve. At the optimized culture medium formula and the experimentalconditions, the OD600value of bifidobacterium breve is about2.6times higher thanthe previous conditions. But it still can not reach the ideal fermentation cultivationexpectation.In the probiotics preparation experiment, the freeze-dried protective formula andfreeze-drying method we chose could make probiotic bacteria survive for threemonths and maintain in the same order of magnitude. Based on the preliminaryoptimized of formula for three different dosage form (granules, tablets and capsules),we obtain good tablets and granular formula, which could satisfy the enteric coat. Thecondition of granules and tablets, such as hardness and brittleness, are qualified. Butthe microcapsule preparation formulation did not achieve the expectation. |
