Penicillin is a kind of key β-lactam antibiotic. Its series of derivatives are the preferred clinical anti-infection agents, so penicillin production constitutes the backbone and foundation of the antibiotic industry. This article aims for improvement the penicillin fermentation level and for reduction the production costs in the scale of industrial product. The optimization of seed develpment process, the substrates concentration control process, the fed-batch process in the scale of120m3fermentation tank, the improvment of fermentation equipments were researched. As the results of our research, the yield of penicillin fermentation product increased by14.1%. The main results are as following:1. Optimization of seed preparation and training process and improvement of the seed quality:Brown sugar substitutes for sucrose as carbon-source in separation medium, and the amount of brown sugar increased by0.3%, the yeast powder reduce to0.2%. The strain grows fast and bacteria colony are thicker, Millet spore incubated time reduce lday, culture cycle shorten1day, the yield of penicillin increases by18%, Improve seed tank inoculation amount of25%, seed incubated time reduce5hours and quality is stable, production fluctuation factor is eliminated because of seed quality.2. Optimization of penicillin fermentation substrate concentration control process and improvement fermentation level:To improve the use of medium effect, citric acid replaces glucose, organic nitrogen-source increases by1%and cottonseed cake powder0.1%in the culture medium, the yield of penicillin increases by5.1%; Supplementary food starch hydrolysis sugar concentration increases by5%, a total of eight tons water is added in later period, and substrate concentration is suitable primary metabolism and the secondary metabolism, the yield of penicillin increases by6.8%, and it is significant for the penicillin production.3.120m3fermentation tank zone put process improvements increase productivity: penicillin fermentation process of training for the release of feed liquid make full use of and to cultivate, To produce more output effect,.Optimization results medium scale20%when put tank titer was higher than11.7%, Optimization medium for corn steep liquor15%, Cottonseed cake powder0.3%, Potassium dihydrogen phosphate0.3%, Ensure belt feeding liquid in the different period of bacteria in the same environment, Keep the fastest growth rate, Produce a greater benefit, Put the tank unit is increased by4.8%.4. Fermentation equipment improvement and reasonable application:fermentation equipment improvement and reasonable application, optimization of agitator combination. Four wide blade propeller agitator and half arc turbine agitator combination instead of double six arrow leaf turbine agitator. Improve dissolved oxygen effect; The converter is used for setting stirring speed. The best setting stirring frequency for:0~30h:40hz;30~100h:45hz;100~160h:50hz;160h~over:45hz. So, the growth of bacteria microenvironment and the fermentation medium dissolved oxygen content improve, but shear force is lower, and it can effectively control the bacteria form; And reduce the tank pressure0.01MPa can reduce fermentation medium carbon content, the yield of penicillin increases by4.1%, and another advantage is reduction of power consumption.
