Tobacco as an important economic crop, has now become an important consumergoods and source of revenue in our country. Tobacco quality is determined by thechemical composition of tobacco, on the premise of smoking is harmful to health, thetobacco industry has been put forward higher requirement on how to effectively reducingtar and harm, and as far as possible to retain the unique fragrance of tobacco. Thus theresearch on the chemicals of tobacco is of great significance for the tobacco quality, theeconomic value of tobacco, as well as reducing tar and harm.Carbohydrate is an important substance in tobacco and its weight accounted for themodulation of tobacco is25%to50%. Meanwhile, it is the structure material of skeletonand network, the carbon frame and energy providers, meanwhile an important factor thataffected the alcohol of the tobacco smoke. In addition, the carbohydrate in appropriatecontent range has an important impact on the aroma and quality of tobacco products,because it can reduce the intensity of tobacco smoke taste, irritation and bitterness, andimprove the aftertaste. Also, there is a certain degree of moisturizing effect for thetobacco products. Therefore, the research on the sugar in tobacco has the reality guidingsense to the development of new cigarette formula and cigarette processing.In this work, carbohydrate (mainly glucose, fructose and sucrose) was selected asthe research object, which has a great influence on cigarette quality.In this work,flue-cured tobacco planting area, a total of30were selected in henan, guizhou, yunnan,fujian four provinces and then the two part of tobacco leaves (C3F and B2F) of eachplanting region was chosen to represent the level of corresponding ones, as well as thetobacco growth year is from2008to2011.In this work, the concentration of glucose,fructose and sucrose in tobacco samples was determined by the high performance ionchromatography using an ion chromatograph (thermo ICS-5000) and sugar ion exchangecolumn (CarboPacPA-20). The test conditions as follows: the column temperature is30℃, benzoic acid and water is used as the mobile phase with the flow rate of0.30ml/min. The reagent preparation and instrument detection is based on the current sugartesting standards in tobacco industry. Moreover, the correlation and difference of glucose,fructose and sucrose content in tobacco samples, including different regions, differentgrowth sites and different growth years, were studied by the different statistical analysismethod of SPSS data analysis software. Also, the data both rated the difference of sugarcontent of tobacco in the relevant condition and the interaction of two factors whether tohave a significant influence on the content of three kinds of sugar. Results in this workprovide a certain objective reference for improving the cigarette production process requirements, selecting the cigarette raw materials and developing new cigarette formula.
