| Sea buckthorn, also known as TFH, is a kind of deciduous shrub belong to Elaeagnaceae Hippophae. Sea buckthorn are mainly distributed in arid zones in China and Shanxi province has the largest sea buckthorn production. Variety of sea buckthorn processing enterprises are in the province, the main products focus on the sea buckthorn extraction of juice squeezing and sea buckthorn oil. Enterprise output a large number of sea buckthorn seed residue in the course of processing, sea buckthorn seeds contain about 30% crude protein, however most of sea buckthorn seed residue are discarded or as feed. The abandoned sea buckthorn seed slags are as raw materials for research in this article, the polypeptide, which have the antibacterial, antioxidant and promoting the growth function were prepared by enzymatic hydrolysis of sea buckthorn seed slags, it provides a new approache to utilize and develop sea buckthorn seeds. The main research contents and progress as follows:1. A study of sea buckthorn seed pre-treatment process. Seabuckthorn seed residue was soaked and then treat with enzyme (cellulase, phytase, tannase), proteins and peptides dissolution rate increased. The results show after the treatment with enzyme hydrolysis, tannins, phytic acid, fiber and other substances had a good degradation, yield of polypeptides increased significantly.2. The hydrolysis technique of seabuckthorn seed residue, sea buckthorn seed residues after pretreatment were hydrolyzed by pepsin, trypsin, papain, acid protease, subtilisin neutral protease B and analyze the hydrolysis effect; the yield of polypeptides were as the indicators to determine the best hydrolysis conditions of different proteases.3. Antioxidant study in vitro of polypeptides hydrolyzed by different enzymes. Antioxidant effect of different polypeptides were compared in vitro and in vivo antioxidant trials. The research shows that the polypeptide hydrolyzed by acid protease has the best antioxidant effect comparing with trypsin, papain in vitro. Using ultrafiltration to make the polypeptide purification, polypeptide molecular weight in the range of 500-2000Da has the best antioxidant effect. With the concentration of polypeptide at 20mg/mL, the eliminating rate of hydroxyl radicals reached to 91.35%, the superoxide anion radical on the removal rate was 64.33%, total antioxidant capacity is 0.906. Polypeptide hydrolyzed by acid protease has good antioxidant effect in vivo antioxidant trials of mice.4. The antibacterial research in vitro about polypeptide hydrolyzed from sea buckthorn seed by enzyme. Using bacteriostatic circle culture method to investigate different proteolytic peptides against E. coli, Salmonella, Staphylococcus aureus and other bacterial inhibition. Tests showed that polypeptide hydrolyzed by neutral protease B has the antibacterial effect to three kinds of bacteria, inhibition zone radii reached 3.3mm,4.2mm and 2.8mm, it’s significantly better than other enzymatic polypeptides. After ultrafiltration, the polypeptide molecular weight in the range of 500 2000Da has the similar antibacterial effect with peptide mixture against pathogenic entero becteria, polypeptides molecular weight in the other range have poor antibacterial effects.5. The study of promoting the growth of peptide hydrolyzed from seed residues by enzyme. The study found that suckling mice feeding peptides hydrolyzed by trypsin feeding for 28 days, the average weight and average height were significantly increased compared with basic group,especially for pups before 14days,the body height,weight and feed efficiency were to promote good effect, but after weaning mice in length and feed efficiency compared with the base group was no significant; auricle separation test, incisor eruption time, long-haired time and our eyes are relatively time and other indicators have significantly advance compared with basic groups; developmental test results like platform righting, hanging off and aerial righting have significantly advance compared with basic groups. Aerial righting hanging drop and the sense of smell to locate have significant differences compared with basic group.It’s indicate that the polypeptide may have a role in promoting growth and development, but need further testing and research. |
