| In order to reveal the effects of season and anti-season tomatoes on contents of the main functional components and physiological function of tomatoes, in this paper, we used season and anti-season tomatoes as the raw materials. The most suitable extraction method and solvent system of carotenoids were choosen, and the HPLC method of carotenoids and cis-isomers were optimized. We also studied the effect of extracts of tomatoes planted in different seasons on improving the D-galactose induced aging mice awareness of injury and oxidative damage. The results were shown as follows: 1 Optimizing extraction method of carotenoidsThree extraction methods and five solvent systems were compared, the best condition was shown as follows: 10 g of the sample was placed in a vessel, wrapping with aluminum foil to be protected from light, and mixed with 100 ml of n-hexane-ethanol(3:4, v/v). The mixture was magnetically stirred with a magnetic stirrer for 30 min. Then 15 ml of distilled water was added to separate the organic layers. The method is simple, less time consuming and highly efficient. 2 HPLC elution conditionsGood separation of 11 kinds of carotenoids including all-trans-forms of lycopene, β-carotene, lutein and 8 kinds of cis-isomers was achieved with a gradient of methyl tert-butyl ether(MTBE) containing 0.05% of TEA(v/v)(A) and acetonitrile containing of 0.05% TEA(v/v)(B) at 1.0m L/min in a 25 min program as follows: 0-8min,0%A-55%A; 8-25 min, 55%A. Detected at 475nm; Chromatographic column is C30. 3 Comparing the carotenoids between Season and Anti-Season tomatoesQualitative analysis showed that the in-season and anti-season tomatoes have the same types of carotenoids, both of them contain 11 kinds of cis- and trans-isomers. But quantitative analysis showed that the content of them exist significant differences. All-trans-lycopene, all-trans-lutein and most of the cis-isomers in Season tomatoes was significantly more than that in Anti-Season tomatoes. but the content of all-trans-β-carotene was significantly lower than that of Anti-Season tomatoes. 4 Antioxidant activity of SF, ASF,SW,ASW in D-gal model in vivo.Mice were injected with D-gal for 6 weeks and were administered SF, ASF, SW, ASW via gastric intubation simultaneously. After six weeks of treatment, the animal behaviors were observed in the open field test and water maze test, and the morpheus of hippocampal and skin were checked. Meanwhile, the activities of antioxidant enzymes(SOD, CAT), non-enzymatic antioxidants, such as the total antioxidant capacity(T-AOC) and glutathione(GSH) level, as well as lipid peroxidation product MDA were evaluated. The results indicated that the SF and ASF attenuated D-gal-induced cognitive impairment via enhancing the antioxidant and preventing the damage of hippocampus. But the effects of SW and ASW was poor. In addition, Season and Anti-Season samples didn’t show significant differences, the Anti-Season tomatoes also possess good antioxidant activity. |
