| Through the optimization of acid bacteria, we screened the bacterial strain of high yield hexanoic acid and acetic acid. Further, the culture medium and culture condition were optimized and lead to product a lot of acid. On this basis, acid esterification was facilitated by adding alcohol and esterification enzyme, and took appropriate distillation method for extraction and concentration, and finally applied to low-grade liquor blending. The main results are as follows:1. The acid producing strains were screened, the acid producing ability was determined and acid distillation methods were compared by experiments. At last, caproic acid bacteria 2# and acetic acid bacteria 2# were determined for producing acid strains and the reduced pressure distillation had higher stripping rate than atmospheric distillation.2. The optimal scheme of the synthesis hexyl ester were determined through the experiments, the system was as follows:2% hexanoic acid,20% ethanol, purified water 78%, esterified starter 25%; esterification conditions were as follows:the esterification temperature 30℃, reaction time 30 days; there was no relation between the effect of esterification and liquid volume and from a practical point of view, the tank should be full.3. The condition of hexyl ester stripping was as follows:the stripping system was comprised by fermentation liquid and edible alcohol which 10% volume of fermentation liquid. The volume of the 10% the reduced pressure distillation method was used and rotary evaporator, distillation pressure was -0.07~-0.08MPa, distillation temperature was 90℃, the speed of 100RPM, and according to the conditions of distillation, the ethyl ester average concentration was up to 2000mg/100mL in the distillate and this was accordance with the content requirements of the blending. The appropriate fermented grains cross-steaming was no influence of hexyl ester content which in the distillate.4. The mix proportion scheme of ethyl acetate was determined through ethyl ester fermentation experiments, specific programs as follows:yeast powder 1%, peptone 2%, D-glucose 4%, zero dosage 4%, pH, fermentation time 20-25days.5. The stripping scheme of ethyl acetate as follows:rotary evaporator and the reduced pressure distillation, the pressure setting -0.07 to -0.08MPa, speed 100rpm, temperature 75℃, liquid direct distillation, the ethyl ester average concentration was up to 2500mg/100mL in the distillate. |
