| In the field of biology, medical treatment and so on, cell injection plays a more and more important role. But there are many deficiencies of manual or semi-automatic manipulation, including low experimental efficiency, difficulty of injection volume control and so on. It is an important subject of cell engineering and mechanical engineering to research a cell injection technology, with the characters of high automation and precise control of microinjection volume.Base on the Digitalization of Micro-Fluids, the technologies of micro devices fabrication, micro- manipulation and microinjection experiments are presented for automatic cell injection in this paper.Based on the thermoforming technology of glass, the fundamental fabrication technologies of cell-microinjection devices are studied. The angle between injection needle and holding needle is resolved effectively using new top-notch and bending technologies.Pressure regulation experiment of cell injection is studied. Then digital micro-injection is realized. Cell auto- transmission experiments are carried out. The influence of driving voltage and frequency on the rate of cell transmission are studied. Then the optimized control parameters are obtained.Micro-needle automatic positioning system is designed. The automation operation of needle insertion and withdrawal are realized. On the basis of pressure regulation, injection experiments are carried out. Quantitative microinjection is realized by selecting the appropriate control parameters.Taking pig oocyte as experimental object, automatic cell injection experiment is carried with experimental efficency of 18 cells per hour. By experimental confirmation, the automatic cell injection is scheme, which is based on cell pressure regulation, cell auto- transmission, micro-needle automatic positioning, and quantitative microinjection. |
