The Chinese natural drugs play an important role in traditional medicine, which are one of the important weapons to cure diseases. They have been applied to the medical treatment for thousands of years. With the development of modern separation technology, the medicinal chemistry has become the hot spot of many scholars. But there were less reports about the properties of metal complexes which were complexed from metal ions and active ingredients extracted from Chinese traditional medicines, especially the reports about the interactions between alkaloids and metal ions.Metal complexes can affect drug physiological activity, significantly improve drug absorption, prolong drug time, and improve biological activity. Metal has synergy to the ligands, and they still can improve efficacy and produce new pharmacological effects. In order to get the drugs with better pharmacological properties, this paper applied calorimeter to study antimicrobial activity.Microcalorimetry was used to study bacterial growth influenced by active ingredients extracted from Chinese traditional medicines and metal complexes. This method can not only provide the proof for the development study but also is a theoretical base for the research of modernization of Chinese traditional medicines.This paper mainly studies the active ingredients of Chinese traditional medicines and metal complex-alkaloids. The main contents are as follows:(1) Active ingredients extract of Chinese traditional medicines.Rhizoma Menispermi alkaloid, oxymatrine and tetrandrine are the active ingredients of rhizoma Menispermi, radix sophorae flavescentis and Radix stephaniae tetrandrae respectively. They were obtained through ethanol reflux extraction, decompression condense, ion exchange and extraction.(2) Synthesis of metal complex-ingredients of Chinese traditional medicines.Extensive experimental results demonstrate that only radix stephaniae tetrandrae in the three alkaloids can coordinate very well with metals. So we synthesized tetrandrine-complexes with iron, cobalt, nickel and zinc firstly. Then characterized the structure by IR and UV spectrum analysis.(3) Studies of bacteriostasis activities.The power-time curves of different concentrations of three kinds of alkaloids and four new metal complexes and M(NO3)2·3H2O were determined by titration microcalorimetry. The growth rate constants(μ) of different concentrations(c) were calculated using logistic equation. The relationship ofμ-c were established and the best inhibitory concentration were obtained. By comparison of their best inhibitory concentrations, the fungicidal activities of new metal complexes on E.coli were better than Tet and M(NO3)2·3H2O.The metal complexes which were firstly complexed from metal ions and active ingredients extracted from Chinese traditional medicines were finally studied on their bacteriostasic actions by microcalorimetry. This is a new idea and research method and is the core and innovation of the thesis. This study can not only provide a basis for screening antibacterial drugs and determining the best amount of drugs but also help the complex research, which involves much knowledge such as pharmaceutical chemistry, biochemistry, chemistry and so on.
