Flavonoid is a kind of polyphenols widely distributed in nature with a wide range of biological activities and pharmacological activity. A large number of studies have shown that flavonoids scavenge free radicals, anti-oxidation, anti-mutagenic, anti-tumor, anti-bacterial, anti-virus, regulating immunity, prevention and treatment hardening of the arteries, reducing blood sugar and other functions. With the development of food industry and consumer attitudes change, natural active ingredients of health food for modern people to pursue the objectives, which flavonoids to pure natural, high activity, quick and role of the characteristics of a wide range gradually being people's attention. The main contents of this paper are as follows:In this paper, a new flavonoid compound chemiluminescence (CL) detection method, and flavonoid compounds, anti-oxidation mechanism of the three studies, the main contents are as follows:1 The determination of flavones with chemiluminescence system of potassium permanganate-glyoxal-sulfuric acidThe determination of flavonoids methods are as follows:spectrophotometry, high performance liquid chromatography, capillary electrophoresis, supercritical fluid chromatography, TLC scanning method. These methods are slow and expensive. There are many merits on CL such as simpler instrumentation without light sources which avoiding the effects of stray light and instability of light source, and thus providing low background with excellent sensitivity, wide linear range, rapidity, simplicity, versatility of use and the easy automation and so on.Through the optimization of experimental conditions, a novel chemiluminescence method for the determination of flavonoids was developed. Flavonoids in acidic conditions, there is a strong sensitizing effect, whereby the establishment of potassium permanganate-glyoxal-sulfuric acid chemiluminescence determination of flavonoids new method. This method has been applied to the determination naringenin, naringin, baicalin, quercetin, puerarin, rutin, luteolin-7-O-galactoside, icariin, genistein more than 9 kinds of flavonoid compounds.2 The determination of flavonoid compounds in three kinds of the mechanism of anti-oxidation by chemiluminescence.Antioxidant activity is the most important biological functions of flavonoids. The determination anti-oxidation of flavonoids commonly used visible light absorption spectrometry, and flow injection chemiluminescence is very few, flow-injection chemiluminescence was uesd to research the antioxidant of flavonoid compounds and compare their activities, and explore the relationship of flavonoid compounds between structure and antioxidant activities.(1)The scavenge of hydrogen peroxideThrough the optimization of experimental conditions, establishment of the luminol-hydrogen peroxide-carbonate buffer chemiluminescence system. Flavonoid and anthraquinone in alkaline conditions inhibit the intensity of chemiluminescence, thus, the new method was established determine the anti-hydrogen peroxide of flavonoid compounds and anthraquinone compounds. This method was used to determination the antioxidant capacities of 18 kinds of flavonoids and anthraquinones, and their IC50 were computed. The results showed that luteolin, rutin, luteolin-7-O-galactoside and quercetin have stronger anti-hydrogen peroxide ability than others.(2) The scavenge of Hydroxyl radicalThrough the optimization of experimental conditions, establishment of the luminol-hydrogen peroxide-copper sulfate-carbonate buffer chemiluminescence system. Flavonoid and anthraquinone in alkaline conditions inhibit the system, thus, the new method was established to determine the anti-Hydroxyl radical of flavonoid compounds and anthraquinone compounds. This method was used to determine the antioxidant capacity of 18 kinds of flavonoids and anthraquinones, and their IC50 were computed. The experimental results show that rutin, puerarin, genistein, baicalin, daidzein, kaempferol, luteolin, luteolin-7-O-galactoside and quercetin have stronger anti-hydroxyl radical ability than others.(3) The scavenge of superoxide anionThrough the optimization of experimental conditions, establishment of a Luminol-Pyrogallol-sodium hydroxide chemiluminescence system. Flavonoid and anthraquinone in alkaline conditions inhibit the system, thus, the new method was established to determine the anti-superoxide anion of flavonoid compounds and anthraquinone compounds. This method was used to determine the antioxidant capacity of 18 kinds of flavonoids and anthraquinones, and their IC50 were computed. The results showed that luteolin, rutin, genistein, luteolin-7-O-galactoside and quercetin have stronger anti-superoxide anion ability than others.These results suggest that:(1) B ring hydroxyl has stronger antioxidant capacity of flavonoid compounds, when there are 3',4'-hydroxy, its antioxidant activity was significantly increased. (2) C ring on the 3-hydroxyl-change will affect the antioxidant effect of flavonoid compounds. (3) 5-hydroxyl groups can enhance the antioxidant activity of flavonoids.(4) In the metal ion-induced free radicals system, C ring of 3-hydroxy effectively can effectively improve the antioxidant capacity of flavonoids, while in the non-metal ion-induced free radicals in the C ring system of 3-hydroxy they contribute very little. (5) C ring 2,3-double bond, and 4-carbonyl conjugation antioxidant activities were not affected of flavonoid compounds. (6) In different anti-oxidant system, the antioxidant capacities of flavonoids are differently, they follow different mechanism of action in different systems.3 The determination of antioxidant activates of people's daily drinking of tea by three kinds of chemiluminescence system.We use the optimized three kinds of chemiluminescence system to determination of antioxidant activates of tea. We optimized extraction process and extraction identification, determination of 46 kinds of tea the capacity of anti-hydrogen peroxide, anti-hydroxyl radical, anti-superoxide anion. The results show that, we can use chemiluminescence to determined antioxidant capacity of tea by the three antioxidant system quickly, and provide useful guidance that select antioxidants from the natural products.
