| P-hydroxybenzoic acid esters (also known as Nipagin esters) is not only widely used in chemical, medicine, food, feed and various industrial corrosion fields, but also used as the cleaning of medical equipment disinfectant, because of its broad antimicrobial spectrum, high efficiency, low toxicity, broad spectrum, easy compatibility.etc. Food preservation and sterilization is the main purpose, it is main preservative to replace sodium benzoate and other food products. At present, the sodium benzoate is still mainly used as preservative in China, but parabens have become one of main presertives because of its toxicity and dosage less than sodium benzoate. Although Nipagin esters are researched and developed later, it become one of hot topics research as preservation and sterilization in China.Many studies confirmed that parabens can not influence uterine weight, the sperm count and spermatozoon morphology of testicular and epididymal, but also have a significant role in tumor cell proliferation, receptor binding assay, recombinant yeast estrogen test in vitro, etc.Drosophila is the most important model organism in biologic study, it has many advantages than other creatures in aging aspect study. In addition, many fruit fly genes has sequence homology with human genes, so Drosophila is usually selected as the material of aging aspects. In our daily life, people can absorb paraben preservatives by variety of ways, and have a certain accumulation in human's body. We inferred paraben have potential hazard on human health through analysing its impact on Drosophila melanogaster. We can protect human health by probing the influence in toxicological and the safety using of comprehensive evaluation. We explored the reproduction and life of Drosophila with PP because BP and PP are permited to use as preservative in Food, the PP can affect the formation of sperm.Here, we studied the survival rate, middle lifespan, mean lifespan, maximum lifespan,90% survival time, developmental cycle, anti-shock capability (including the capacity of anti-hunger, and heat shock and climbing skills), the activity of superoxied dismutase (SOD) and the content of alondiadehyed (MDA) of different days in male and female, and also observed the bodyweight of Fi generation during development changes in five different connditions of PP (the control group:0.00 %, low PP groups:0.02% and 0.05%, high PP groups:0.10% and 0.20%). At last, in order to explord its toxic, we analysed the change of aging and reproduction of Drosophila, and also clarify the relationship between the both. The results show:1. The reproduction capacity of male and female of parental flies can be reduced in low and high concentrations of PP, the reproduction can be increaseed fertility in low concentration and significantly decreased in high concentration when their F1 generation are given the same treatment.2. Low concentration 0.02%,0.05% PP and high concentration 0.20% PP reduced the survival rate, middle lifespan, mean lifespan, maximum lifespan of male and female, whereas the various indicators of lifespan in the 0.10% PP was significantly higher than others. The results show that the PP can effect the lifespan of Drosophila.3. Comparaed with the control group, the capacity of anti-hunger, anti-heat shock and climbing skills of male and female in low concentration PP groups were not changed. High concentration 0.20 % PP accelerated the aging process, and reduced the capacity of anti-hunger, anti-heat shock and climbing skills of male and female, whereas the climbing ability of flies can be increaced in the high concentration 0.10% PP.4. The SOD expression of female and male of Drosophila in the low PP groups were fewer than the control group, the high concentration 0.10% PP improved the SOD expression of Drosophila, whereas 0.20% PP reduced the expression of SOD in male and female. The MDA content of low PP groups and the 0.20% PP group were higher than the control group throughout of the development cycle., whereas its content in 0.10% PP was increased more slowly.5. The body weight of F1 female generation was enhanced in low concentration 0.05% PP group, whereas the body weight of all F1 male and female generation in different concentration of PP groups were lower than the control group. With the increasing of PP concentration, the body weight were decreased.6. Low concentration PP groups can reduced the body weight of F1 male generation, whreras high concentration PP groups were slowed down the rate of growth and development of male and female offspring at the same time. We summarize that PP can effect the growth cycle of Drosophila. |
