Surface Plasmon Resonance technology is a new type of photoelectric detection. Since the first application of the surface plasmon resonance (SPR) phenomenon for sensing almost two decades ago, this method has made great stides both in term of instrumentation development and application. It has the characteristics of high sensitivity, high resolution and anti-electromagnetic interference. Its tesing process is fast. Able to get the actual data and it is easy to operate, no need to mark and maintain the biological activity of molecular and so on. It is suitable for the detection of the trace substance. Since Nylander and Liedberg used the SPR-sensing technology for gas detection and biological sensors for the first time in 1982, SPR-sensing technology has been more and more widely used in optics, chemistry, biology, environment, and food analysis, medical and pharmaceutical fields. Now, SPR sensor technology has been commercialized and SPR biosensors have become a central tool for characterizing and quantifying biomolecular interactions. Their implementations, instrumentation development and application also get the rapid expansion of the development.A novel measurement approach of Surface Plasmon Resonance (SPR) with bi-beam was presented in order to improve the sensitivity and anti-disturbance capability of intensity-modulated SPR sensor. Because the excitation of SPR phenomenon is mainly P-ray, the front of the light source must put a polarizer. On this basis, this article proposes a new idea without a polarizer. And we compare P-ray and PS-ray in the method of emulation to get a feasible basis in the theory. Then we verify its feasibility through experiment. This method greatly simplifies the previous experimental apparatus.
