Glucosinolates are found in cruciferous plants a wide range of content of secondary metabolites. Glucosinolates and their defense in plant-derived products, human nutrition and cancer has a special role.Now generally accepted glucosinolates biosynthesis of the model includes three main stages:(1)side-chain extension (2)glucosinolates core synthesis (3)the modification of the second side-chain. The three stages of this biosynthesis-related genes and enzyme characteristics has been detected in Arabidopsis.This experiment on behalf of select Brassica plant broccoli, rape, cabbage, Chinese cabbage for the test materials, the core of glucosinolates synthesis pathway for the key ACGM marker gene CYP79A2. Brassica choose as subjects not only because of Brassica vegetables containing glucosinolates and generally rich in content and, more importantly, Brassica vegetables widely cultivated in China and has considerable economic value.According to this experiment have been identified in Arabidopsis thaliana glucosinolate structure of the CYP79A2 gene sequence of the conservative design of specific primer, and to select the material of total genomic DNA as a template for PCR reaction, and PCR products were sequenced and analyzed. Gene expression in order to overcome the problem of low abundance, and increased use of DNA template, reducing the use of primers, appropriately increasing the annealing temperature, the pilot repeatedly.Analysis of PCR products in the distribution of the genus Brassica, an important metabolic pathway through a comparative analysis of genes to explore the various types of Brassica genetic relationship between, and may be modified for the use of metabolic engineering of crops Brassica Glucosinolates of metabolism to provide a means of molecular-assisted selection.
