HSL plays an important role in lipid and energy metabolism.Many research about HSL-deficient mice showed the male HSL knock out mice are infertile. These mice have decreased testis weights and testicular anormalities,including the presence of multinucleated round and elongation spermatids,vacuolization of the seminiferous epithelium,asynchronization of the spermatogenic cycle,and a marked reduction in the numbers of late spermatids.These results show that HSL play an important role in spermatogenesis.The lack of HSL has an effect on the spermatogenesis and leads to the infertility of male.Isolation and identification of spermatogenesis related proteins and further functional study are very important to the elucidation of molecular mechanism involving in meiosis and spermiogenesis.It is also important to clinical diagnosis and treatment of male infertility and male molecular contraception.The aim of our test is trying to explore the reason of infertility in Hormone-Sensitive Lipase(HSL) gene knock out mice.For these studies we utilized a proteomic approach with two-dimensional gel electrophoresis(2-DE) analyses and mass spectrometry(MS) identification of proteins with altered expression in mouse testes between normal and HSL gene knock.The HSL+/+ and -/- mouse we chosen have same mother and father.With analyzing and using of 2-DE technique,hundreds of spots were isolated per gel.In these processes,we used different ways,and discuss the benefits of dissimilar methods.
