Debaryomyces sp. was screened from 19 strains of microorganisms, which produced glycerol kinase (GK). The optimization of cultivation conditions was selected through fermentation tests. The highest enzyme activity was obtained through 25h cultivate at 31℃ and 160 r/min, when 120ml of the culture in 250ml shake flasks was grown in lg/L glucose, 3g/L yeast extract, 6.0g/L citric acid, 1.8g/L KC1, 0.5% glycerol. The initial pH was adjusted to 5.0. The mocuiums volume was 6.0% (V/V). Under the optimized cultivation conditions, the enzyme activity reached 1.19U/mg and the thalli productivity reached 18g/L.
