AP-1 (The activator protein 1), as a very important cell biosensor, can response to a plethora of external stimuli and regulate a wide range of cellular processes. It is a homo- or hetero-dimers composed by Fos , Jun , ATF and MAF protein family members and belongs to basic region-leucine zipper (bZIP) transcription factor family. Fos and Jun are the most common AP-1 proteins in mammalian cells with c-Fos and c-Jun dimmer being the main form due to its stability. AP-1 binds to TPA response elemet (TRE:5' >TGAC/GTCA<3' )and regulates the downstream genes expression in response to growth factors, cytokines, UV, stress, viral and bacterial infections and carcinogen etc.AP-1 which locat in the end of cascade of signal pathway is a bridge of external stimulus and cell function. The genes induced by AP-1 are diverse in multiple functions such as proliferation, differentiation, transformation, apoptosis , inflammation, survival or death , and immunity etc. Therefore, finding the positive stimulus of AP-1 will be beneficial to anti-tumor , anti-inflammation and anti-apoptosis et al, and still helpful to elucidate the mechemism of growth.Our work is aimed to construct a stable cell line of AP-1 signaling pathway to screen any stimulus activating the signal pathway and analyze the drug efficacy and specificity associated with AP-1 . The positive stimulus can be used as a drug candidate for further clinical trial. Our stable cell line was derived from human 293E cells with chromosomal integration of an anti-puromycin gene and a GFP reporter gene construct regulated by 8 copies of the AP-1 response element that we synthesized in the company.
