In order to generate the expected phenotype properties, the transgenic plants usually need the exogentic gene high expression in the particular space and time. The promoter takes a key effect on decision of the gene expression. In the present work, the seed-specific promoter LegA gene is choosen to construct a set of 5' deletion expression vectors named leg0029I, leg0029III, leg0179I, leg0179III. This work will give some information and work as control for the following research of the new seed specific promoter of Mabinlin II gene in our group.Arabidopsis is named the "drosophila"of the plant. It is always regarded as the model experimental material of the traditional genetics and the molecular biology. In the recent years, it becomes the first material of the growthing genetics. In order to keep up the step with the internation, ararabidopsis has been introducted into china for the experimental material. In our laboratory, the applation of the arabidopsis is limited as the variety of climate, temparature and humidity, et al. In order to sufficiently use the model plant, it is very important to be in proficient in the cultivating technology. According to the experimental conditions of our laboratory and the climate characters, the experiment made the preliminary searches on the cultivation medium, seed gemmation, watering, humidity, light condition and the two common transgenic methods of arabidopsis (Vacuum infiltration method and Dip method). In this paper, I have acquired some data on arabidopsis successful cultivation and transformation. Meanwhile, I transformed some expressed vectors to the arabidopsis and have obtained some transgenic arabidopsis from selection medium. This work will offer the utilitarian experience for the application of the model plant-arabidopsis in our laboratory.
