The article mainly studied three aspects: The effect of Glycerol on survival of E.coli implanted by low energy ion beam, transferring the E.coli induced by Ion beam ,and constructive improvement in the methods of ion-beam microbe technology.The study of the effect of Glycerol on survival of the E.coli shew Glycerol could protect E.coli and improve dry, vacuum and implantational survival rate during ion beam implanting the E.coli. The whole survival rate of protected the E.coli implanted by 30keV N+ was 530-10000 times higher than unprotect ones. Glycerol changed bacteria outer color and survival rate curve. At the same doses, the color of the E.coli protected by five and one percent Glycerol was white or yellow, lighter than unprotected. In addition, the survival rate curve of the E.coli with Glycerol located above one without Glycerol, and the peak of which was higher and delayed at dose.Three kind of ion beam, N, Ar and Mg ion beam, and nine doses , 1x1014N/cm2, 2x1014N/cm2, 3x1014N/cm2,5x1014N/cra2,1x1015N/cm2,2x1015N/cm2, 3x1015N/cm2, 5X1015N/cm2, 1X1016N/cm2, were applied at transformation induced by ion beam. Among 130 experiments ion beam inducing plasmid DNA directly transfer the E.coli, 8 of which succeeded. Although transformational rate is low, some law were found: Ar ion beam was superior to N and Mg ion beam; The dose 1-3 X 1015N/cm2 was more help for ion-beam transformation; For plasmid DNA, superhelix DNA was more efficient than lined DNA and 20 u g/ml DNA was better concentration . Those conclusions gave valued reference for microbe ion-beam transformation.Addition iron ion solution to E.coli implanted by low energy ion beam can improve transformation rate. Especially, the effect of the bivalent Ca2+ and Mg2+ solution was several dozen or hundred times as high as the one of univalent K+ and Li+. For K+ and Li+, 4mol/L K+ and 2mol/L Li+ is more efficient than 0.2mol/L K+ and 0.2mol/L Li+. In a word, Cooperation of the ion beam implantation and iron ion solution promoted plasmid DNA transform the E.coli.On the foundation of practice and work, a series of prompt and simple methods was set up,such as method spreading bacteria on small glass slices and on eatable filmand scrubbing bacteria form glass slices.
