Taxol, a diterpenoid compound, is able to bind and stabilize microtubules, blocking cell replication in the late G2-M phase of cell cycle and resulting in cancer cell withering. So it is a potent anticancer drug. This paper study the effect of carbon source, precursor (Phenylalanine, Tyrosine, ammonium acetate) and hydrogen peroxide on biomass, taxol yield of Y3 which is Endophytic taxol-producing fungi of plant. Moreover, The taxol of zymolytic extraction is analyzed by Thin layer chromatography (TLC), High performance liquid chromatography (HPLC), Micellar electrokinetic chromatography and MALDI-TOF.The results show that the optimal carbon source of Y3 is glucose and the optimal concentration of Phenylalanine, Tyrosine, ammonium acetate is respectively 100mg/L, 9mg/L and 20mg/L; Hydrogen peroxide can distinctly promote the producing of taxol. Under the condition of addition concentration of Hydrogen peroxide is 8mmol/L every 8 hours, the yield of taxol get to 700ug/L, over 9 times higher than the contrast.What's more, The identification of the fungi strain has been made, and Y3 is Rhizopus arrhizus Fischer which is the first reported Endophytic taxol-producing fungi of plant at home and abroad.
