Gamma-aminobutyric acid (GABA) receptors are the G-protein-coupled receptors for GABA, the main inhibitory neurotransmitter in the mammalian central nervous system. GABAB receptors are unusual in that the homologous GABAB1 and GABAB2 subunits need to heterodimerise in order to reach the cell surface.Lipid rafts are specialised microenvironments within biological membranes that are enriched in cholesterol and sphingolipids. Lipid raft domains are platforms for plasma membrane signalling complexes. They are key regulators of G protein coupled receptor function. The G protein coupledĪ³-aminobutyric acid receptor B (GABAB receptor) co-isolates with lipid raft domains from rat brain cerebellum. So in this paper,we investigated that the regulation of rafts for lateral diffusion of GABAB receptor by Totle internal reflection fluorescence microscopy in transfected HEK293 cells.So in this paper,we investigated that the regulation of rafts for lateral mobility of GABAB receptor by Total internal reflection fluorescence microscopy in transfected HEK293 cells.We discovered that GABAB2 receptor regulated the translocate mobility of GABAB1 receptor,while the mobility of GABAB1 mutant, which was able to atomatic ally to the cell membrane with a radom trajectory. Was not regulated by GABAB2 according to sigle-molecule imaging. Lipid raft domains are platforms for plasma membrane signalling complexes. We found that the diffusion coefficient of GABAB receptor heterodimers increase when cholesterol is distrupted,and the lateral mobility of GABAB receptor is regulated by lipid raft. Compared to GABAB1a receptor, GABAB1b receptor is easier to translocate lipid to the lipid raft.When the cells were treated with MCD,we found that Median Instantaneous Diffusion Coefficient of GABAB1b-containing heterodimers is higher than that of GABAB1a -containing receptor.This paper only a first investigate for lateral mobility of GABAB receptor and are underpinned for researching of molecular mechanism.
