| 1. Study of the Extraction of Flos Carthami(1) Study of the extraction: With solvent volume and extracting time as examing factors, using the transferring rate of hydroxysafflor yellow A as index, we selected the optimum extraction condition by Uniform design, which the method was: boiling 2 times,60minã€50min respectively, with water of 12ã€10 times of Flos Carthami.(2) Study of purificationThe contrast test showed that the B was the best macroporous adsorptive resins for the absorbtion and separation of Flos Carthami. The optimal parameters of purification for Flos Carthami were sample concentration (1:2.5), sample pH (2), sample flow velocity (2BV/h), then being eluted by water (1BV, pH=2),50%EtOH(6BV) at the velocity of 5BV/h, gathering the part elued by 50%EtOH.2. Study of the Extraction of Rhizoma Chuanxiong (1) Study of the extraction:Contrast experiment showed that water was a better extract solvent for Rhizoma Chuanxiong. With the transferring rate of Chuanxiongzine as index, the optimum extraction condition was selected by orthogonal test. Results:Taking water as solvent, Rhizoma Chuanxiong was extracted three times with 8,6 and 6 times volume of water for 1.5h,1.0h,1.0h, respectively.(2) Study of purificationThe contrast test showed that the B was the best macroporous adsorptive resins for the absorbtion and separation of Rhizoma Chuanxiong. The optimal parameters of purification for Rhizoma Chuanxiong were sample concentration(1.5:1), sample pH(4), sample flow velocity (2BV/h), then being eluted by water(2BV),50%EtOH(4BV) at the velocity of 5BV/h,gathering the part elued by 50%EtOH.3. Study of Quality StandardThe content of Chuanxiongzine, Ferulic acid, hydroxysafflor yellow A were determined by HPLC method. The content of Safflor yellow was determined by UV.The methods above are precise, reliable and repeatable and can be applied for the content of the Extraction of Flos Carthami and the Extraction of Rhizoma Chuanxiong. |
