| This paper mainly analysis the enzymatic hydrolysis of Ophiopogon polysaccharides, the monosaccharide composition, and the hydrolyze ability of Absidia sp.O8s. For the comprehensive utilization of Ophiopogon, the paper has done a correlation analysis to sapions.In this paper, Ophiopogon crude polysaccharides were separated and purified by Sephadex G-100. Then its composition would be analyzed by TLC and HPLC. The results showed that the monosaccharide was mainly composed of fructose and glucose. Further quantitative analysis by HPLC in20μL of acid solution, fructose concentration is0.80mg/mL, glucose concentration is0.066mg/mL. Thus Basic compositions of sugar moiety in Ophiopogon polysaccharides were fructose and glucose, and molar ratio between fructose and glucose was approximately12:1.The results showed that the enzymatic hydrolyze of Ophiopogon polysaccharides is fructose. Also the enzyme from Absidia sp.O8s has the ability of cutting the multi-glycosidic bond, making the macromolecules polysaccharide to smaller molecules.The method of viscosity determines the different materials to the molecular weight. It shows that the average molecular weight of Ophiopogon polysaccharide is26K, alkali leaching of Ophiopogon polysaccharide molecular weight is19K, the enzyme hydrolysis products of the molecular weight is16K.Through the weighing comparative, water extraction of crude saponins is32.9%and alkali extraction is35.66%. If by AB-8purification, water extraction of the total saponins is0.40%and alkali extraction is0.82%. If by Butanol extraction, water extraction of the total saponins is0.62%, and alkali extraction is1.33%. |
