Heterocyclic aromatic amine was mutagenic/carcinogenic compounds formed in the process of meat products which had rich protein. In order to human health, it was vital to establish a stable and accurate detection method for HAAs. HAAs are present at trace quantities (ng g-1level) in complex matrices, and a high number of matrix interferences can be present, so before using apparatus detected HAAs, we should conduct preparation and clean-up procedures. In this paper, we choose five typical HAAs as object of study, was developed via a new synthesized materials solid-phase extraction coupled with high performance liquid chromatography-ultraviolet detection system for the detection of the five HAAs in meat products.According to the structure of the five HAAs, a new materials as enrichment sorbent was prepared in the form of bulk polymerization which use MAA and4-VP as functional monomers, EGDMA as cross-linking agent, AIBN as initiator and both of DMSO and Tol as special porogen. Through characterized by FT-IR, SEM and evaluated by adsorption experiments, the materials exhibited appropriate internal structures and revealed high adsorption capacity and fast kinetics ability for adsorption of heterocyclic aromatic amines. The five HAAs (10mg L-1) could reach at more than85%of equilibrium adsorption capacity within5min and the adsorption equilibrium is reached within30min.Using prepared adsorbent as the HAAs SPE materials, A new offline SPE coupled with HPLC-UV method was established and used in the determination of five HAAs through optimizing the conditions of solid phase extraction such as sample types and elution solvent, the volume of eluent etc. Under the optimal experimental conditions, the results reveal a good linearity of the five heterocyclic aromatic amines with correlation coefficient (R2)>0.9959were achieved over a ranging of2.5ng L-100μg L-1, the detected limits(S/N=3) were ranging from0.3μg L-1to0.6μg L-1. The peak area precision (RSD) of peaks area for eleven parallel determinations was under6%, the spiked and recycling experiment exhibited the average recoveries (n=3) was ranging from99.31%to106.45%.In this study, in order to verify the accuracy and practicability of the developed method, we applied the developed method into the detection of HAAs existing in cooked pork, chicken and grilled fish which exposed to person. The sample of spiked and recycling experiment with three concentration(4μg kg-1,10μg kg-1,20μg kg-1) of the five HAAs exhibited the average recoveries was ranging from68.21%to115.37%and the RSD(n=3) were less than10%. The experimental results showed that the developed method was practicability, could be used for analysis of HAAs in meat products. |
