Actinidia can improve digestive function of stomach and intestines. It is reported that Actinidia peel and pomace contain plenty of dietary fibers; solubility dietary fiber-pectin (SDF-P), specially, which is condensated from galacturonic acid (D-GA). SDF-P has strong water-adsorption capacity in the digestive tract, then it can increase the volume of intestinal and promote intestinal peristalsis, which may improve elder gastrointestinal functions, reduce or prevent the intestinal disorders and the incidence of colorectal cancer. Previous study showed that acetylcholine M receptor (Ach-M) distributed in gastrointestinal mucosa and smooth muscle cells. When the receptor is activated, secretion of digestive juices accompanied by bowel movements is up-regulated. In our study, we used Actinidia peel and pomace as materials to established a triple-craft process on the key cellulase accompanied with HCl acid and microwave treatment for degradation of cell wall of the plant. Furthermore, we investigated mechanism of pectin effects on intestinal epithelial cells and smooth cell muscle cells with extracted and purified solution of SDF-P.1Establishment of effective extraction and purification of Actinidia SDF-P:Three main factors, as enzyme or acid concentration, solid to liquid ratio and microwave time, were found with dose screening. Cellulose of peel and pomace is hydrolysed by HCl to cleave the conjunction of macromolecules, then treated with cellulase and microwave to form a triple-craft process so that effectively controlling enzymatic hydrolysis rate and molecular weight and promoting the yield and purity of SDF-P. The yield in peel and pomace is8.96%and6.67%, the purity is up to64.82%±0.1736%and52.84%±0.0572%, respectively. Compared with single HCl treatment or enzyme hydrolysis, the yield is increased2-11times and purity increased3-16times. Our data showed pection content in peel is higher than it in pomace. This triple-craft process based on cellulose reduced processing part significantly, which can effectively improve the productivity and directly used for industrial production.2Research for mechanism of pectin effects on primary smooth cell muscle cells (ISM), Caco-2cells and DLD-1cells with extracted and purified solution of SDF-P. We found10μM SDF-P could induced contraction of cell skeleton, Ca2+influx and up-regulated concentration. Under this SDF-P dose, AchM2in Caco-2cells and DLD-1cells internalized and gathered to cell nucleus. Furthermore, Western Blot demonstrated β-arrestin is colocalized with AchM2in Caco-2cells and ISM cells. We presume that Actinidia SDF-P has been similar function to Ach, promoting AchM2receptor internalization mediated by β-arrestin to enhance smooth muscle contraction. Our study verified Actinidia SDF-P can promote intestine smooth cells contraction and epithelial cells motility, and it is a hopeful dietary supplements to improve intestinal function.
