| Kandelia candel is a common mangrove plants mainly distributed in the bay area of the muddy tidal flats, and periodic suffer water inundated in the intertidal environment. Zhejiang province is the most northern ares (28°25’N) of the geographical distribution of mangrove plants in China. At present, there are only a few researches on the bioacitive components from mangrove plant’s in Zhejiang province.Kandelia candel, which produced in Zhejiang province Wenzhou City Yueqing Coastal wetland, is our subject’s principal research object. There are two main aspects about this project.First, establish a determination methods of flavonoids, and then optimize the extraction technology of flavonoids.(1) With NaNO2-Al (NO3)3method, we explore the content of the total flavonoids. Through the methodology experiment, quercetin shows good linear relationship in the range of1.35-8.10μg/mL, its regression equation is y=0.053x-0.003(R2=0.9990), its recovery is98.75%-102.50%, RSD=0.30%(n=5). Therefore this method is accurate, and we could use it testing the flavonoids content in the leaves of Kandelia candel.(2) With NaNO2-Al (NO3)3method, we explore the extraction of the total flavonoids from Kandelia candel. Through the single factor experiment, we explore four factors which are temperature, ethanol concentration, material liquid ratio and time on the influence of flavonoids extraction yield; Through L9(34) orthogonal experiment design, we explore the best extract technology of flavonoids. Two experimental results are basically the same, the optimal extraction process was:80%alcohol, liquid ratio1:30,90℃,1hour for backflow. Impact factors:material liquid ratio> alcohol concentration> time> temperature.Second, explore the best extract technology of antioxidant activity.The experiment method by DPPH method,2-deoxy-D-ribose method, Fe3+reduction experiment, we evaluated the antioxidant activity of in different angles, through comparing the two different types of extract technology, we can judge the antioxidant activity and flavonoids whether they have correlation between them.(1) DPPH method:80%alcohol, liquid ratiol:35,90℃,1hour for backflow. Through this method we get the sample, and it shows strong clear ability among the others. Although the date shows the sample is weaker, the change of colour is obvious, which we can see it has strong ability to clare DPPH free radicals.(2)2-deoxy-D-ribose method:80%alcohol, liquid ratio1:25,80℃,1hour for backflow. Through this method we get the sample, and it shows strong clear ability among BHA and the others.(3) Fe3+reduction:80%alcohol, liquid ratio1:25,90℃,1hour for backflow. Through this method we get the sample, and it shows strong clear ability among the others.But it shows weaker ability than the BHA.To sum up, we have not found the correlation between the antioxidant activity and flavonoids, the antioxidant activity extract technology is different from the flavonoid extract technology. So we should take the other factors to evaluate. |
