Pigments are more and more widely used in the food industry and rapid detection of synthetic pigment has become a research hotspot in recent years. Today, more and more research points to the consumption of synthetic pigments on the body side effects such as carcinogenic, teratogenic, mutagenic and other side effects.It has been obvious that tartraine and sunset yellow can cause hyperactivity and affect children’s mental development.It is urgent and necessary to improve fast detection methods for synthetic pigments.This study established a HPLC method for simultaneous determination with a Gaussian software.A brief and accurate method was developed to simultaneously determine nine legal synthetic dyes (New Red、Tartrazibe、Amaranth、Indigo Carmine、Ponceau4RC、Sunset Yellow、Allura Red、Brilliant Blue FCF、Erythrosine) in food by high performance liquid chromatography with diode array (HPLC-DAD). The developed chromatographic method employed an Gemini C18column (250mm;4.6mm;5μm). Mobile phase consisted of A:20mM ammonium acetate and B:methanol. A gradient elution program was used in this work. Detection wavelengths were the optimum absorption wavelength of each dyes. New Red was detected under507nm,Tartrazine was under426nm, Amaranth was under521nm, Indigo Carmine was under611nm, Ponceau4RC was under509nm, Sunset Yellow was under483nm,Allura Red was under508nm, Brilliant Blue FCF was under629nm and Erythrosine was under535nm. Column temperature and flow rate were kept as35℃and1.0ml/min respectively. Injection volume.was10μL. Higher limits of detection(0.006-0.026mg/L) and quantification (0.020-0.142mg/L) together with satisfactory recoveries were obtained. The proposed method can be applied to successfully screening of nine legal synthetic dyes in simple food system.Nine pigments’ mid-infrared spectras were obtained by Fourier transform infrared spectrometer. Nine pigments’ predicted mid-infrared spectras were obtained by using Gaussian09W software. Differences of the predicted spectras among nine pigments were analyzed and it were pushed to the real spectrum so that I can find the differences among the real spectrum.It was used to supplyment the qualitative analysis of nine pigments.
