| Bacteria from fresh Paralichthys Lethostigma skin were seperated and purified, using a combination method of the traditional and modern automated equipment identification. The analysis of bacterial flora was completed in this research. The results showed Gram-positive bacteria strains accounted for18.9%of the total strains, including Staphylococcus aureus strains of12.6%, Micrococcus strains of6.3%;Gram-negative bacteria accounted for72.3%of the total strains, including advantaged strains of Enterobacter(l9A%), Liquefaciens (10.7%), Moraxella(8.3%), Vibrio(7.8%), Alcaligenes(73%) and Shewanella puterfaciens (6.8%) among206seperated bacteria. The results indicated that there are a large number of Enterobacter in fish skin, the culture water are polluted. It provides the optimal theoretical basis to study the specific spoilage organism from frozen Paralichthys Lethostigma.In the paper, the bacterial flora composition of4℃and0℃frozen Paralichthys Lethostigma were identified in the qualitative and quantitative analysis as the same method. At the same time, comparing with sensory indicators, microbial indicators, nutrition indicators, freshness indicators and chemical indicators, specific spoilage organisms were confirmed in the frozen on many views of the shelf life. The experimental results indicated that the specific spoilage organisms were confirmed as Pseudomonas spp.and Shewanella puterfaciens at4℃and0℃. The proportion of Pseudomonas spp.and Shewanella puterfaciens to the total corrupt bacteria was93%and81.2%; TVBN (Total Volatile Base Nitrogen) were66.92mg/100g and70.45mg/100g respectively; pH values were7.87and7.60. When cultured Paralichthys Lethostigma was storaged at0℃and4℃, the shelf life were12days and8days respectively.It is significant to study bacterial flora of Paralichthys Lethostigma for which can provide theoretical foundation and scientific basis to find out the health status of farming environment, confirm the corresponding improvement measures, take further effective measures for the preservation, optimize the frozen process of fresh Paralichthys Lethostigma and develop new technology. |
