| Herba Epimedii was Chinese traditional medicine and reputed to reinfore the kidney yang,strengthen the tendons and bones,and relieved rheumatic condition,Epimedium flavonoids and their bioactivity were researched mostly.Recently researches indicated,blood of human body was absorbable for secondary glycosides or aglycones.In this paper,the hydrolysate produced by hydrolyzing technology of acid and refing and separating technology for secondary glycosides and aglycones were attempted.The total epimedium flavanoids and icariin had been separated from Epimedium extract by means of column chromatography which absorbed with macroporous resin HPD100and polyamide eluted with ethanol/water system. The ingredient of total epimedium flavanoids had been charactered by means of HPLC-ESI/MS.When epimedium flavanoids were split by ESI/MS.Glycons of the C7were split after Glycons of the C3in the positive mode.But condition was opposite in the negative mode.The H2O was probably bond to Glycons of C7position.But final conclusion would be certificated by next test.The hydrolysate of total epimedium flavanoids and icariin were analyzed by HPLC-PDA-UV.The hydrolytic conditions and effecting facter of hydrolyzing for epimedium flavanoids had been summarized.Solvent,acid concentration and reaction time played an important role in the hydrolyzing reaction.Cariside I (purity90%) would be produced from icariin when concentration of hydrochloric acid was0.5mol·L-1and hydrolytic reaction time was0.5h.Anhydroicaritin (purity90%) would be produced from icariin when concentration of methanol of acid was2.5mol·L-1and hydrolytic reaction time was5h.The hydrolysate mixtures which hydrolyzed by hydrochloric acid from total epimedium flavanoids included five kinds of compounds.five kinds of compounds were separated and purified by column chromatogramphy from this mixtures.Chemcial constitutions of them were identified by mass spectrum.five kinds of compounds included secondary glycosides and aglycor. ne secondary glycosides were caohuodise D and platy petaloside A,The aglycones were icaritin, brevicornin and anhydroicaritin。 When epimedium flavaniods had been hydrolyzed by hydrochloric acid,Glycons of the C7were removed after Glycons of the C3.The epimedium flavaniods before hydrolyzing were mostly8-isopentene radix flavaniods.Hydrolysate included8-isopentene radix and8-isoprenylkaempferol radix epimedium flavaniods.This paper presumed the hydrolyzing law of epimedium flavaniods by hydrochloric acid.But next test would be necessity. |
