Obeject:To design and synthesize multi-pyridine ligands and corresponding copper complexes. Using the synergy of metal ions and ligand, do screening for the discovery of new copper complexes with multi-pyridine ligands which have good bioactivities and magnetic properties.Methods:According to the current literature, we have designed and synthesized the new multi-pyridine ligands and their copper complexes through several methods; The structures of these complexes have been determined by X-ray diffraction, Element analysis and IR measurement; We also investigated the interaction of these complexes with DNA by fluorescent spectra and gel electrophoresis. Adopting MTT method, we survey the anti-tumor activity of these complexes; In addition, we investigated magnetism of these complexes by variable temperature magnetic susceptibility and variable field magnetic susceptibility measurements.Result:We successfully synthesize three multi-pyridine ligands and five new copper complexes and characterize their structure through several kind of measure method; Using spectrofluorometric method, complex2has been proved with good DNA combining activity. Kapp (complex2)=1.80×105M-1. Gel electrophoresis results show that complex1and complex2exhibit chemical nuclease activities in the presence of H2O2. The result of cytotoxicity experiment displays that complex1and2are dose-dependent. For HeLa cell line, the IC50(48h) of complex1is2.16±0.04μM, complex2is4.24±0.03μM, which show that complex possess good ati-tumor activity. Magnetic studies reveal that the complex3has strong intrachain ferromagnetic interactions between the Cu(Ⅱ) ions.Conclusion:Through synthesis and characterization, we successfully get three new complexes and their crystal structures; We discuss bioactivity of complexes by chemistry, molecular biology and cell biology related means, the result indicate that complex1and complex2possess good biological activity. These complexes have potentialities to act as new antitumor agents, and the further study on the relevant mechanism is underway. In addition, complex3shows strong ferromagnetic interactions, which may be used as molecule-based magnets.
