In order to reflect the true composition of volatile components in the sample, andimprove the accuracy and reliability of Mass Spectrometry. In this thesis, thermal desorptioncombine with gas chromatographic and mass spectrum for the first time was applied toanalyze aroma components of fresh corns, and MassworksTMsoftware combine with spectrallibrary search for qualitative analysis of the composition of angelica oil components. Thefollowing conclusions:⑴The results showed that thermal desorption’s optimal parameters for analyzing aromacomponents of corn were30℃adsorption temperature,2h adsorption time and-30℃coldtrap temperature.38kinds of aroma components in corn had been detected in two testedsamples of ordinary sweet corn and purple waxy corn,34kinds of them were detected inordinary sweet corn and30kinds in purple waxy corn. The relative contents of the commoncomponents in the two corns reach80.223%and90.783%respectively. Ordinary sweet cornhas8species of endemic components and the relative content of16.042%, purple waxy cornhas4species of endemic components and the relative content of7.998%. This method ofanalyzing volatile components is simple, green, good repeatability, high efficiency enrichment,and the volatile components were measured near the olfactory sense of smell, which are thereal components of sample. It is an effective pretreatment technology for extracting volatilecomponent.⑵The volatile components in angelica oil from MINXIAN were analyzed for the firsttime by gas chromatography-quadrupole mass spectrometry and gas chromatography-ion trapmass spectrometry. Total of thirty-three compounds were identified by MassworksTMsoftwareanalysis, MS/MS qualitative analysis approach of GC-IT MS and library search. The majorvolatile components of essential oil from angelica are Z-Ligustilide, E-Ligustilide, β-Ocimene,α-Pinene,3-Butylidene phthalide and γ-Elemene. In addition to, Nerolidol,Globulol and Eudesmol are first isolated from the volatile oil of MINXIAN. Results showedthat the results obtained by three kinds of qualitative methods which are MassworksTMsoftware analysis, MS/MS qualitative analysis approach of GC-IT MS and library searchmutual authentication could improve the accuracy and reliability of identification of unknowncomposition.⑶MassworksTMsoftware and NIST library identified the formula C15H24of naturalisomeric substances that were α-Pinene, Sabinene,β-Pinene,β-Myrcene,β-Ocimene,α-Ocimene.and the formula C10H16of natural isomeric substances that wereβ-cedrene,β-Farnesene,γ-Curjunene, Chamigrene,γ-Elemene, Cedarene,β-bisabolene, Himachalene.
