In this work,systematic studies were carried out on optimize the extractionconditions of yellow pigment; analysis of volatile components of dried flowers;isolation,purification,identification of yellow pigments from Leucosceptrum canumSmith; identify its safety through the toxicology experiment.(1) Based on single factor tests, the optimum extraction conditions ofmicrowave-assisted ethanol technology of yellow pigments from Leucosceptrumcanum Smith are obtained by the response surface method. The results showed thatthe optimum extraction conditions were as follows: alcohol concentration70%(V/V),microwave power350W, microwave time80s, the predicted value of extractionefficiency of yellow pigments from was95.23%,and it was preferably fitting withthe predictive value of98.38%.(2) With the increase of market share of natural pigment, the separation andpurification of natural pigment process must keep pace with the times, so chooseusing membrane separation technology to separation and purification the yellowpigment from Leucosceptrum canum Smith. In order to ensure the combination ofmicrofiltration, ultrafiltration and nanofiltration to purification yellow figment, socompares the influence of coarse filtration, ultrafiltration, nanofiltration on theextracts of yellow pigment from Leucosceptrum canum Smith. The results show thatthough using the combination of microfiltration, ultrafiltration and nanofiltration toprocessing crude extract, can retain yellow pigment solution with fewer impurities,color value was7.82(1%,1cm,431nm), thus purify the pigment.(3) Yellow pigment compounds2was isolated and purified by the traditionalseparation methods or membrane separation from Leucosceptrum canum Smith, andthe laboratory animal were KN mouse(SPF grade). Toxicological research includedacute toxicity test (Horn, LD50), mouse bone marrow micronucleus test, mouse spermmalformation test and salmonella typhimurium/mammals microsomal enzyme test(Ames test). The results showed that: the LD50value of compounds2was more than21.5g/kg, The results of the sperm chromosomal aberration test and the bone marrow micronucleus rates were negative, no mutation effect for body cell and reproductivecell was observed at dose of2.50~10.0g/kg, Ames test was also negative. Socompounds2was classified as non-toxic grade.
