Natural bone is composed of mineralized collagen fiber with hierarchicalstructure. The development of mineralized collagen was supposed to “be tuned byorganic matrix” according to the present studies. However, there is no authoritativetheory about the principle of mineralization about bone, and the process also isdifficult to mimick in vitro. Research on the biomimetic construction of mineralizedcollagen systematically will benefit to grasp the mechanism of biomineralization, alsobenefit to the development of the repairing material and regeneration material ofbone.In the study, the orderly template for mineralization self-assembled withself-made type I collagen from rat-tail was to establish. Self-made EMPs (EnamelMatrix Proteins) from pig, DMP1(Dentin Matrix Protein1), PAA (PolyAcrylic Acid),TPP (Tripolyphosphate) and β-GP (Beta-glycerophosphate) were used as inducingfactors, and SBF (Simulated Body Fluid) and PBS (Phosphate BufferSolution)-CaCl2solution worked as biomimetic solution. The experiments ofbiomimetic construct were carried out under37.5℃. POM, AFM, FSEM were used toobserve the microstructure of the products and XRD characterized the phase species.Under the tune of PAA, a hierarchical structure from nanoscale to micro-scalecomposed by the ordered mineralized collgen fiber was obtained, also together withqusia-bone period ribbon construct in the collagen fiber. The mineralized collagenfibres kept highly ordered structure with unified size even in the microscale level,which verified the mechanism of PILP (Polymer Inducing Liquid Process) on boneformation in some degree.TPP and β-GP had strong ability to inducing the formation of nucleator in thebiomimetic solution, while its ability to tuning collagen mineralization was suffered tolimited. So, when TPP and β-GP worked as inducing factors, mineralized collgencomposite fibre still can keep its ordered structure in nanosacle level. Beingbiomacromolecules, EMPs and DMP1show a strong ability to forming nucleator. The formed microcrystals and EMPs were easy to composite with collgen fibre, whichconstructed an ordered mineralized structure in a higher level. It is imply that therewere interaction between biomacromolecle and collagen.The novelty of the study is to improve the hierarchical structure from nanoscaleto microscale level in viro via biomimetic constructing in an ordered collagentemplate.
