Capillary electrophoresis with electrochemical detection (CE-ED) has beenstudied more than ten years. Compared with other separation technology, CE-ED isstill in the early stage, especially for the application in real samples. Owing to theadvantages of high efficiency, sensitive, and low sample and sovlent consumption, thedevelopment and application of this technique become more and more extensive.CE-ED has been widely used not only in life science, but also in the field ofenvironment and food analysis. To solve the food safety and environmental pollutionproblems, the quickly and accurately detection of toxic and harmful contaminants hasbecome more and more important.In this thesis, new CE-AD (amperometric detection) methods were established forthe separation and determination of chiral drugs clenbuterol, phenolic environmentalestrogens and sugar, sugar alcohols, coupling with on line concentration techniques.Four parts are included in this thesis. In the first part, the development and theprinciple of CE were introduced. An overview on applications of capillaryelectrophoresis especially in sugar, environmental estrogens and chiral drugs ispresented. The developments and the prospects of capillary electrophoresis aresummarized. Finally, research topics are presented in this thesis.In second part, a CE-AD method using hydroxypropyl-β-CD (HP-β-CD) as thechiral selector was successfully developed for the enantioseparation and determinationof clenbutetol. And the inclusion mechanism and the separation mechanism of chiralanalytes were studied. Under the optimum conditions,(±) clenbuterol were baselinesepatated with the detection limit of1.03ng/mL. Linear range were2.5~400ng/mL.Eventually, the proposed method has been successfully applied for the analysis ofhuman urine sample.In the third part, CE-AD method was successfully developed for the separationand determination of three important phenolic hormone(BPA, DCP, PCP) inenvironmental water samples. With methanol as additive, and the effect of methanolon the apparent electrophoretic mobility and the peak shape of analytes was studied.Under the optimum conditions, three analytes were separated completely within14min. The detection limits of BPA, DCP and PCP were0.068μg/mL,0.041μg/mL and 0.24μg/mL, fulfilling the requirement of national minimum concentration standardof sewage discharge.In the last part, in extreme pH conditions, a CE-AD method was developed forthe separation and determination of sugar and sugar alcohols in tobacco sample. Thatprovides an efficient, simple, reliable method for indication of tobacco product quality.This method has been successfully applied in tobacco samples with the recoveryranged from82%~105%, RSD<7.0%.
