| Marbofloxacin, which is only used in animals, is a new kind of fluoroquinolones with chemical name:9-fluoro-2,3-dihydro-3-methyl-10-(4-methyl-l-piperazinyl)-7-oxo-7H-pyrido[3,2,1-ij]-1,3,4-benzoxadiazine-6-carboxylic acid. Many researches have been done about it on pharmaceutical kinetin. And the research reports show that it has a great prospective market.Taking2,3,4,5-Tetrafluorobenzoic acid as a raw material, the goal product was obtained through tow methods. And the reaction conditions were optimized. The total yields were3.7%. The products were confirmed by melting point and1HMR.In this article a new synthesis method of marbofloxacin was studied on. As6and7of ethyl6,7,8-thrifluoro-1,4-dihydro-1-(N-methyl-formamido-)-4-oxo-3-quinoline carboxylate can all react with4-methyl-piperazine, we take cyclization before condensation with N-methyl-piperazine, which obtained the goal product. No6substitution by-product was found by HPLC and1HMR. The total yield was18.68%taking2,3,4,5-tetrafluorobenzoic acid as a raw material.The results showed that the process of synthesis of marbofloxacin can be realized with more simple operation and the higher yield can be gained, which is superior to that of the reported one. |
