| With the development of molecular biology and cell biology, genetically modifiedanimals have been successfully established. The safety of genetically modified (GM) hasaroused the attention from various circles of society. The safety of GM food has greatimportant meaning for development of transgenic technology, promotion of geneticallymodified foods and even the problems of food shortages.According to the conception of Substantial equivalence issued by FAO/WHO,theprevious research of the assessment for GM plant and the national standard for the safety offood, we designed the procedure for GM milk assessment. Through the compositionalanalysis, bioinformatics analysis and the30-day feeding study, we have assessed the safety ofGM milk. Basis on the experiment and the statistics we could provide some foundation forlate applications of GM milk and offer some explorations for building the system for GMfood safety assessment.1. Compositional analysis of milkThe human β-defensin-3(HBD3) milk was used as the sample and the traditional milkwas used as blank control in this study. Assessment contains the basic nutrition(total protein,fat, lactose, pH, nonfat milk solids), assessment of amino acid, assessment of mineralelement(Caã€Mgã€Pã€Kã€Naã€Feã€Znã€Se), assessment of vitamin(Va Ve Vc Vb1Vb2). TheresultS show that the GM milk has no significant difference from the traditional milk. Somelittle differences has no statistically significance.2. Bioinformatics analysisUsing the Pepetide Mass, ProtParam tool and Protscale provided by the website ofExPASy, we commenced the physical-chemical analysis and3D structure analysis of HBD3.Using the SDAP, Farrp, PDB and PHYRE, we analyzed the homology sequence andhomology3D-structure of allergenic proteins, and predicted the allergenic of HBD3. Resultsshowed:1) the HBD3is not from the eight-type allergens.2) The physical-chemical analysis,indicated thatHBD3is an unstable protein.3) Bioinformatics analysis showed the HBD3doesnot possess the sequence or3D-structure homology with the well-known allergenic proteins.3. Thirty-day safety study of GM milk on Kunming mice This experiment has divided into three groups: GM milk groups, tradition milk groupsand blank control. After30-day feeding, we anaesthetized the mice to take the blood. Theblood was used for analysis ofhematological and serum chemical parameters. Then, we didrough anatomy and picked out the primary organs for pathological examination. Resultsshowed that no difference was found among in all those hematological and serum chemicalparameters, and no pathological changed was observed in all groups. |
