Quinazoline and its’ derivatives have wide applications for their importantbiological activity; at the same time, quinazoline derivatives are the basic skeleton ofmany natural products, which provide a train of thoughts for the molecular design andsynthesis of medicine, pesticide and materials. Although many methods for preparingquinazoline derivatives are available, but there remain some disadvantages, such aslong reaction time, high temperature, and requirement of special catalyst, etc.Therefore, some novel, efficient, environmental benign and easily-handed synthesisroutes need to be explored. In this paper, quinazoline-4-tosyl-ates were used asstarting reactants to prepare two types of potential biological activity quinazolinederivatives.Firstly, we have developed a novel catalytic system to implement the couplingreaction of quinazoline-4-tosyl-ates with cyano group by utilizingDPPF/Pd(OAc)2/Cs2CO3as catalyst, at the temperature of80centigrade in toluene. Itcan furnish a rapid and straightforward access to4-CN functionalized quinazolinederivatives that are potential biologically and medically useful compounds. Theresults indicated that under this catalytic system all kinds of substrates can obtainmoderate to good yields. At the same time, we have illustrated the leaving ability of4-methylsulfonate ester and the other halogens.Secondly, we have achieved azidation of quinazoline-4-tosyl-ates undercatalyst-free conditions. The results showed that sodium azide is a good azidationreagent for quinazoline-4-tosyl-ates azidation. Under optimal conditions, a series ofpotential biologically substances of4-azide quinazolines derivatives were generated.The reaction was green and environmental friendly, with convenient operation, whichprovided an efficient method for construction of a small molecule compound libraryof4-azide quinazolines derivatives.
